In this paper,new indocyanine fluorescent probes A-1,N-1,M-1 and Q-1 were synthesized using indocyanines as reporter groups through condensation,substitution and Acylation reactions.The test shows that probes A-1,N-1,M-1 and Q-1 have strong UV absorption and fluorescence spectral response to biothiols,and they also obtained good results in the detection of endogenous/exogenous biothiols in the Hep G2 cells:(1)Among them,the maximum UV absorption wavelength of probe Q-1 is 620 nm,and all exhibit a ratiometric fluorescence response to biothiols;(2)Under physiological conditions,they have good stability,of which probe M-1 has good stability in a wide p H range;(3)The probes have good selectivity for biothiol and are not interfered by other amino acids and inorganic cations;(4)Kinetic studies have shown that all probes except probe A-1can fully respond within 30 minutes and remain unchanged;(5)Cytotoxicity experiments show that the toxicity of the probes to cells are almost negligible;(6)Probe Q-1,N-1 have good fluorescence signal in Hep G2 cell fluorescence imaging.Studies have shown that the probe shows good selectivity and sensitivity to biothiols.The probe can be used for quantitative analysis of endogenous and exogenous biothiols.
