ObjectiveRenal ischemia-reperfusion injury(IRI)usually occurs after trauma or surgery,leading to renal failure and poor clinical prognosis.In the present study,we aimed to use bioinformatics analysis to identify central genes,potential transcription factors during renal IRI pathology,predict potential therapeutic targets and drugs for patients with renal IRI.MethodsBy Robuss Rank Aggreg(RRA)analysis,using GSE39548 ? GSE58438 ?GSE71647 ? GSE87024 ? GSE131454 and GSE148420 from Gene Expression Omnibus(GEO)database to identify differentially expressed genes(DEGs)between renal IRI group and control group.The enrichment function and pathway of DEGs were analyzed by GO and KEGG analysis.According to the results of MCODE and cell tumor,the central genes were obtained by Protein-protein interaction(PPI)network analyst.GSE98622 and HK-2 cells were used to verify the expression of central genes.TRRUSTv2 database was used to identify potential transcription factors in the kidney.Finally,we predicted a small molecule drug TWS-119,which might be a potential drug for renal IRI.ResultsIdentification of 12 central genes,included Jun?Cxcl1?Cxcl2?Il1b?Ccl2?Icam1?Serpina1d?Spp1?Il6?Rcn1?Timp1?Lgals1.GSE98622 and q RT-PCR results showed that the expression of these genes was also significantly increased in addition to renal tissue and HK-2 cells.Jun?Nfkb1?Cebpb?Sp1?Egr1 were the most important transcription factor associated with renal IRI.TWS-119 might be a potential drug for renal IRI.Conclusion(1)The DEGs closely related to renal IRI are mainly focused on TNF signaling pathway,MAPK signaling pathway,NOD-like receptor signaling pathway,Toll-like receptor signaling pathway and HIF-1 signaling pathway.(2)The 12 hub genes associated with renal IRI are Jun?Cxcl1?Cxcl2?Il1b?Ccl2?Icam1?Serpina1d?Spp1?Il6?Rcn1?Timp1?Lgals1.The expression of hub genes in RNA-seq data and cell levels showed statistically significant.The 5 key transcription factors were Jun?Nfkb1?Cebpb?Sp1 and Egr1.(3)TWS-119 minght be the potential small molecule drug for the treatment of renal IRI.
