Diet,as an essential life activity in human life,has an important influence on the structure of gut microbial community composition.In recent years,there are more and more studies focusing on gut microbial,most of which collect some fecal samples at a certain time point to represent all intestinal microflora,while few reports pay attention to the daily changes of gut microbial community of volunteers.Therefore,this study analyzed the differences of short-term gut microbial of young people under normal diet,and further explored whether different sampling methods affect the gut microbial of people.This study is of great significance to broaden the research of gut microbiota in human health and other fields.In this study,the diversity of microorganisms in fecal samples of 10 volunteers at different time in their daily life was studied by metagenomic sequencing technology combined with multivariate statistical methods,and the short-term gut microbial differences of young people under normal diet were revealed.The results showed that:(1)Comparative analysis showed that there was no significant difference in the overall structure of species in feces collected by "local sampling" and "all sampling"(P-value > 0.05),and most species had no significant difference between the two groups(P-value > 0.05).Only low abundance species such as Atopobium parvulum,Bacteroides faecis and Prevotella buccalis had significant differences between the two groups(P-value < 0.05).While,the same species and function in stool samples of the two groups should a significantly positive correlated(R = 0.72,R = 0.94,P-value <0.05),and the microflora structure and function modules of the two groups were also highly consistent(P-value < 0.001).Therefore,"local sampling" can meet the needs of gut microbial research.(2)The study on the dynamic changes of gut microbiota of volunteers showed that the dominant bacteria in the intestines of volunteers were Firmicutes,Bacteroidetes,Proteobacteria and Actinobacteria.The dominant strains were Eubacterium rectale,Faecalibacterium prausnitzii,Bacteroides vulgatus,etc.There were great differences in the composition of gut microbial among different volunteers,but in a short time,most dominant species had no significant changes in the intestinal(P-value > 0.05).(3)There were few kinds of stable bacteria in young people's intestines,but their relative abundance were high,which can perform most of the functions of gut microbiota.Bacteria belonging to Bacteroidetes play an important role in maintaining the stability of gut microbiota structure of young people.In daily life,the structure and function of young people's gut microbiota have always maintained dynamic stability in a short time.Therefore,collecting fecal samples within a certain time will not have a great impact on the research results of gut microbiota,and can still meet the requirements of experimental analysis.On the basis of evaluating the influence of sampling methods on the results of young people's gut microbiota by using metagenome technology,this study focused on analyzing the dynamic changes of young people's gut microbiota and function under17-day normal diet.In this study,it's confirmed that the structure of gut microbiota of volunteers changed dynamically in daily life,but collecting fecal samples in a certain time had no significant effect on the overall structure of gut microbiota,which also laid a theoretical foundation for large-scale population experiments.
