Objective: To investigate the impacts of high-fat diet on body length,body weight and learning and memory of PS offspring rats in prenatal stress experiments.To analyze the pathophysiological effects of GluA1 on the physical development and learning and memory of offspring rats.Methods: Pregnant rats were divided into four groups: control group,PS group,HFD group and PS+HFD group.PS group: pregnant rats were given restraint stress during 14-20 days of pregnancy,3 times each day and 45 minutes each time;HFD group: fed HFD;PS+HFD group: given PS and fed HFD.21 days after birth,the offspring rats were divided into control group,PS group,HFD,PS+HFD group according to gender(n=8).The body length and body weight were measured to observe the effects of HFD on the growth and development of offspring rats.The Morris water maze test was used to observe the effects of HFD on the learning and memory of offspring rats.The expression of GluA1 protein in hippocampus of offspring rats was detected by Western blot.Results: The body length of male and female HFD groups was significantly higher than control groups(P<0.01),but the body length of male PS+HFD group was significantly lower than HFD group(P<0.01).The difference of body lenthe and weight didn't find in female.The body weight of male and female HFD group was significantly higher than control group,but the body weight of male PS+HFD group was significantly lower than HFD group(P<0.01);Compared with male PS+HFD group,the weight growth rate of male HFD group was significantly higher(P<0.05).The difference of body weight growth rate didn't find in female.HFD could significantly increase the latency of PS offspring(P<0.05).PS could reduce the expression of GluA1 protein in hippocampus of offspring rats(P<0.05),while HFD could reduce the expression of GluA1 protein in hippocampus of offspring rats(P<0.05).Conclusion: HFD could promote the weight growth of normal male and female offspring rats,but its effect was inhibited by PS in male offspring rats,it didn't affect female offspring rats.HFD could significantly reduce the learning and memory ability of offspring rats induced by PS,which the underlying mechnishm might be the decrease of GluA1 protein expression in hippocampus of rats.
