Polarization spectral imaging technology,as a new optical detection technology that combines spectral imaging and polarization detection,has been extensively used in many fields.Polarization is part of the important information of light.When light passes through normal cells and cancerous cells respectively,the polarization information contained in it is changed by the diverse influences of the two types of cells.In this paper,polarization detection and spectral imaging technology are used to obtain polarization information images,to study the difference between polarization images of normal cells and cancerous cells,and distinguish between the two types of cells.In this study,a microscopic polarization spectrum imaging system based on Acoustooptic Tunable Filter(AOTF)and Liquid Crystal Variable Retarder(LCVR)was built.A parameter calibration scheme for LCVR based on AOTF spectroscopy was proposed,and the relationship between the incident light wavelength,phase delay and driving voltage was determined.A verification experimental light path with linear polarizer as the measured object is established,which confirmed the correctness of the experimental light path.Taking biological tissue slices as the experimental target,combined with opencv image processing method based on python language,all Stokes parameter images of tissue slices were obtained,which indicated that the method could extract Stokes parameters normally.With normal ovarian cells and cancerous ovarian cells as the final experimental subjects,all the Stokes parameter images of the two types of cells in a series of light wavelengths were successfully obtained by the above method.In order to obtain clearer experimental results for comparative analysis,the image algorithm was strengthened,and Sobel edge detection was utilized to optimize experimental imaging results.The comparison between the original Stokes parameter image and the Stokes parameter image extracted by Sobel edge proves that the image processing algorithm is feasible to improve the imaging quality.Differences of Stokes parameter images of normal and cancerous ovarian cells were analyzed,which provided a new idea for the application of polarization spectroscopy in the detection of cancer in the medical field.
