As a kind of motor protein,kinesin combines and hydrolyzes ATP,causes the conformation change of neck,and urges the exoprotein to "walk" along the microtubule to complete the transportation of goods in the cell.Since the driven protein is discovered,researchers have studied their working mechanism from many ways and have made many progress.In terms of experimental research,a series of characteristics such as stepping mode,run length distribution,motion speed,lateral movement along the axial movement of the microtube;various theoretical models of driving protein axial motion mechanism were established in theoretical aspects.Successfully explained some experimental phenomena.However,for the study of drive protein lateral movement,less experimental data has less,and the relevant theoretical model has not been established.Based on the following two considerations,this paper attempts to establish a theoretical model of lateral movement of kinesin.Firstly,there are many potential obstacles in the process of kinesin transporting goods on the complex and crowded microtubule channel in the cell.In order to maintain the two-way flow of goods,kinesin must effectively avoid the obstacles on the way forward,and lateral movement should be a way to bypass the obstacles.Secondly,the established model can explain the long-distance directional transport along the microtubule move.According to the structure of microtubule and the movement characteristics of kinesin,combined with the existing mechanism of axial movement of kinesin,the "random walk" model and "central gravity" model of lateral movement of kinesin are proposed.According to the established model and experimental data,the running length distribution and velocity of kinesin were calculated.The theoretical results show that the calculated results based on the "central gravity" model are basically consistent with the experimental data,and the "central gravity" model is more suitable to describe the lateral movement of kinesin than the "random walk" model.This thesis is divided into four parts.The first chapter mainly introduces the kinesin family and the structure and function of microtubules and the transport mechanism of kinesins;the second chapter introduces some experimental results and theoretical models of kinesins,focusing on kinesins The third chapter is the main part of the paper.Based on experimental phenomena,this article proposes several types of kinesin lateral motion models,and calculates,analyzes,compares and discusses the rationality of various models.The fourth chapter is the summary and prospect of the main work.
