Jujube(Ziziphus jujuba Mill.)is native plant in china and is a jujube plant of the Rhamnaceae.As the main variety of red jujube in northern Shaanxi.Jujube cultivation is concentrated in the Yellow River basin and Wuding River basin.It is revealed by high-throughput sequencing analysis of the rhizosphere soil samples and field soil samples of Jujube in the four counties of Yulin and Yan'an.The microbial diversity in the rhizosphere of Jujube in northern Shaanxi.High-throughput sequencing analysis of the jujube rhizosphere soil samples and field soil samples of the four main jujube producing counties in Yulin and Yan'an reveals the rhizosphere microbial diversity of jujube in northern Shaanxi;the matrix method is used to collect jujube roots GC-MS analysis of its composition is the basis for revealing the root exudates of Jujube;the use of coating method combined with functional medium is used to screen out the plant growth-promoting rhizobacteria of Jujube rhizosphere,and the compound application of different rhizobacteria is carried out to explore the preliminary results.The growth-promoting function is expected to screen out the compound microbial agent suitable for Shaanbei Jujube.High-throughput sequencing analysis results show that at the phylum level,the dominant species are Actinobacteria,Firmicutes;at the class level,the dominant species are Actinobacteria,Bacilli,and.Alphaproteobacteria;dominant species Micrococcales and Lactobacillales at the order level;dominant species at the family level are Vicinamibacteraceae,Pseudomonadaceae,Streptomycetaceae;at the genus level The dominant species above are Pseudomonas and Bacillus.The root exudates of sterile tissue cultured Jujube seedlings were collected by the matrix culture method,and the composition of the root exudates of Jujube was identified by GC-MS analysis.Among them are allelochemicals: isoamyl caproate,phenol,vanillin,2,4-di-tert-butylphenol,2,6-Di-tert-butyl-4-methylphenol,2-methoxy-4-(methoxymethy)phenol,dimethy 4,5-imidazole dicarboxylate,phenyl salicylate,dibutyl phthalate,2,5-bis(1,1-dimethylethyl)-Phenol,docosenamide,3-mercaptohexanol butyrate,5-methyl-thiazole,ethyl propionate,1,2,3-trimethyl-benzene,chlorodibromoacetic acid,bis(2-ethylhexyl)phthalate,butylatedhudroxyluene,1,2,3,4-tetramethylbenzene,ethyl4-t-butylbenzoate,5-hexenoicacid,2,6-dimethoxyphenol,4-methyl-1,2-benzenediol,catechol,methoxyphenol,1,4-dimethyl-piperazine,3-aminoisoxazole and 2-amino-benzoxazal-2-Alanine.Through the dilution and coating culture of rhizosphere soil and the functional medium optimization,two plant growth-promoting bacteria Bacillus halotolerans JT-25 and Bacillus cereus JT-86 that can be used in compound microbial agent were identified.Compared with the control group,JT-25+JT-86 treatment can promote the increase of wheat shoot length,root length,fresh weight and dry weight;JT-25+JT-86 treatment can significantly promote soil peroxidation than the control group hydrogenase activity increased significantly,soil urease and soil invertase activity increased significantly,soil alkaline phosphatase also showed differences compared with the control group;JT-25+JT-86 treatment can significantly increase the chlorophyll content and peroxidase activity significantly reduces wheat malondialdehyde and free proline content.
