Abstract/Summary:
Aedes albopictus is one of the top 100 invasive animal species in the world.It is one of the most important vectors of vector-borne diseases for prevention and control in China.Dengue fever is the most important mosquito-borne infectious diseases transmitted by Ae.albopictus,mainly caused by Dengue virus(DENV).DENV is divided into four serotypes,among them,DENV2 is the most transmissible.As there are no effective vaccines and therapeutic drugs for dengue fever,prevention and control of its vector mosquitoes is the main method.However,due to the long-term and largescale use of chemical insecticides,the resistance of mosquitoes has increased year by year.There is an urgent need to find new and environmentally friendly mosquito control technologies.Microbial control have become a research focus in recent years.This study combines metagenomic sequencing and bioinformatics analysis methods to carry out two researches on the microbiome of Ae.albopictus: The first study is the microbiome related to mosquitoes infected with DENV2.The three groups of mosquitoes that were infected with viral blood meals,those that were not infected with viral blood meals,and those who consumed non-toxic blood meals were respectively subjected to metagenomic sequencing to detect the differences in the microbiome composition and abundance of the three groups of mosquitoes.Microorganisms related to mosquito infection with DENV2 have been screened out.The second study is the microbiome related to mosquitoes sucking blood meal.Through metagenomic sequencing of mosquitoes fed sugar water and blood meal,the differences in microbiome composition and abundance of the two groups of mosquitoes were detected.The types of microorganisms associated with mosquitoes sucking blood meals were screened out.The results are as follows:1.The dominant phyla of the microbiome of Ae.albopictus are Proteobacteria and Ascomycota,and the dominant genera are Aspergillus and Metarhizium.2.Through screening of microorganisms related to DENV2 infection by Ae.albopictus,15 different microorganisms were found,including Alphaproteobacteria bacterium 65-37,Methyloglobulus morosus,Shigella sonnei,etc.The results of microbial function analysis showed that their corresponding functions were all functional substances such as proteins and enzymes.They are basically life-related enzymes and proteins involved in circulation,protein synthesis,and material decomposition.These microorganisms may be involved in the immunity and metabolism of mosquitoes,and the different susceptibility of mosquitoes may be related to the different content of these microorganisms in their bodies.It is speculated that if artificial interventions are made on the content and activity of these microorganisms in mosquitoes,it may change the susceptibility of mosquitoes to viruses.3.Through the screening of microorganisms related to different foods(blood meals and sugar water)in Ae.albopictus,28 different microorganisms were found,such as Lachancea mirantina,Rhizobium yanglingense,Bacteroides luti,etc.Analysis of the KEGG pathway showed that there are differences in functions such as amino acid metabolism,lipid metabolism,replication and repair,translation,etc.,indicating that after mosquitoes take a blood meal,related microorganisms will improve the metabolism of mosquitoes and help mosquitoes to develop their ovaries to better complete reproduction process.By intervening and regulating these microorganisms,the normal physiological activities of mosquitoes can be disrupted and the mosquitoes cannot reproduce normally,so as to achieve the purpose of controlling mosquito populations.Since the microbiome of mosquitoes with different virus susceptibility will have corresponding differences,this study firstly aims to understand the differences in the microbiome of mosquitoes with different susceptibility,and to establish the relationship between the mosquito microbiome and susceptibility.It is expected that artificial intervention in the microbiome will indirectly affect the susceptibility of mosquitoes,thereby reducing the spread of mosquito-borne diseases.Since the microbiome of mosquitoes that feed on blood or on water will have corresponding differences,and mosquitoes need to feed on blood to reproduce,this study aims to understand the differences in the microbiome of mosquitoes that feed on different foods,and to establish the relationship between the microbiome of mosquitoes and different foods.It is expected that artificial intervention in the microbiome will indirectly affect the blood-sucking behavior and physiological changes of mosquitoes,thereby reducing mosquito reproductive ability then reducing the number of mosquito populations.
