In recent years,due to the gradual improvement of people's living standards,the requirement for livestock meat has gradually increased,so the animal husbandry has also increased.However,when raising cattle and sheep,if a great quantity of cattle and sheep manure in the farm can not be treated in time and effectively,a large number of toxic and harmful gases will be produced in the farm.The most common and harmful gases in cattle and sheep feces are NH3 and H2S.If these harmful gases are not treated in time and effectively,cattle and sheep will suffer from many diseases for a long time,which will not only affect the health of cattle and sheep,but also directly influence the quality and texture of meat.At the same time,people's long-term absorption of these gases will also harm people's health and cause many related diseases.Therefore,it has become the top priority of animal husbandry to restrain the release of odor gas from cattle and sheep feces.The deodorization method of microecological preparation refers to the method of adding foreign microorganisms to the faeces of cattle and sheep to make their microorganisms react with the odor gas produced by the faeces of cattle and sheep,degrade and utilize NH3 and H2S,and reduce the emission of odor gas.This method not only has low cost,but also can inhibit the emission of odor gas on the source head,so it becomes the best way to deal with animal faeces in animal husbandry Method.Four dominant Deodorizing Bacteria WZ1,WZ2,WZ4 and JDF1 were screened out from ten well preserved strains in the laboratory,and their biological identification was simple.The culture conditions of dominant deodorizing bacteria were optimized.Finally,the dominant deodorizing bacteria were applied to sheep feces to study the deodorization effect.1.Screening of deodorant bacteria from sheep manure.Ten strains of bacteria,KC1,CB1,JDF1,MM1,JM1,WZ1,WZ2,WZ3,WZ4 and WZ5,which are preserved in the laboratory,will be screened out,and the ten strains will be cultured and inoculated into fresh sheep feces,so that they can fully react in the gas absorption device.The release amount of NH3 in feces was measured by NH3 absorption titration with boric acid.The dominant deodorizing bacteria were WZ1,WZ2 and WZ4,which could inhibit the concentration of NH3 to 69%,61%and68%.The quantity of H2S released from feces was surveyed by methylene blue spectrophotometry.The dominant deodorizing bacteria were WZ2,WZ4 and JDF1,which can inhibit the concentration of H2S to 96%,93%and 90%.Strains WZ1,WZ2,WZ4 and JDF1were selected as high efficiency deodorizing strains.2.Optimization of culture conditions of Deodorizing Bacteria in sheep manure.In this experiment,the dominant deodorizing bacteria were inoculated into the basic culture medium of bacteria,and the conditions of the selected deodorizing bacteria were optimized by shaking table culture and fermentation.The optimized factors were respectively culture p H,culture temperature and shaking table revolution.Each factor was set at five levels,and single factor experiments were conducted respectively.Finally,the best foster conditions were resolved by722 spectrophotometer.The results displayed that the best foster condition of WZ1 was p H7.5,temperature 33?,revolution 140r/min;the best foster condition of WZ2 was p H6.5,temperature 33?,revolution 160r/min;the best foster condition of WZ4 was p H6.5,temperature 30?,revolution 180r/min;the best foster condition of JDF1 was p H6.5,temperature 36?,revolution 200r/min?3.Study on the application of Deodorizing Bacteria in sheep manure.Four dominant deodorizing bacteria were screened out from the feces of sheep,and then they were cultured in the basic medium of bacteria with the most suitable culture conditions,and applied to the fresh feces of sheep for comparison with the blank control group to get the application research of deodorization effect.The results showed that the NH3 reduction rate of WZ1 was69%when the inoculation amount was 8 m L.When the inoculation amount of WZ2 was 6m L,the NH3 reduction rate was 61%.When the inoculation amount of WZ4 was 8m L,the NH3decreased by 68%;when the inoculation amount of WZ2 was 2m L,the H2S decreased by96%.When the inoculation amount of WZ4 was 8m L,the reduction rate of H2S was 93%.When the inoculation amount of JDF1 was 6m L,the reduction rate of H2S was 90%.Compared with the blank control group,the concentration of NH3 in the dominant bacteria which inhibited the concentration of NH3 was reduced to some extent.The effect of inhibiting the concentration of NH3was WZ1>WZ2>WZ4.The effect of inhibiting the concentration of H2S in the dominant bacteria which inhibited the concentration of H2S was WZ1>WZ2>WZ4 Compared with the blank control group,the concentration of H2S in the dominant deodorizing bacteria decreased in varying degrees,and the effect of inhibiting the concentration of H2S was WZ2>WZ4>JDF1.Through the application of the experimental results,it can be proved that the selected dominant deodorizing bacteria have obvious inhibition on NH3 and H2S in sheep manure.
