Drug resistance is one of the most intractable issues in current clinical treatment of cancer.The heterogeneity of tumor cells and microenvironment adaptations to targeted therapy can promote the acquisition of drug resistance and the metastasis of cancer.The inherent heterogeneity of cancer cell population is that it contains drug-sensitive and drug-resistant cells.In targeted therapy,drug-sensitive cancer cells secrete multiple soluble factors into the tumor microenvironment.These factors enhance the growth,dissemination and metastasis of drug-resistant cancer cells and support the survival of drug-sensitive cells.Therefore,microenvironmental adaptation plays an important role in the rapid emergence of acquired drug resistance.Evaluating cancer therapeutics in the context of tumor heterogeneity and microenvironmental adaptation is very difficult.In this paper,the complex interaction mechanism is simplified into a five-dimensional dynamic model.This article is divided into three parts,which mainly study drug resistance due to treatment.The research results can help people a deeper understanding of resistance mechanisms and provide a strong scientific basis for clinical treatment.In Chapter 1,we briefly introduce the biological background of tumor drug resistance,tumor growth model,basis model of cancer drug resistance,pharmacokinetics model and the basic theories of ordinary differential equations.In Chapter 2,we mainly present the mathematical model of therapy-induced cancer resistance.This model is based on cell dynamics,pharmacokinetics,and micro environmental adaptability to describe the interactions among sensitive cancer cells(CS),drug-resistant cancer cells(CR),metastatic cancer cells(CM),angiogenic cells(CK)and drug concentration(D).In this chapter,the existence and stability of the non-negative equilibrium points of the model are analyzed and discussed in detail,and the existence of each positive equilibrium point is simulated.In addition,we also analyzed the influence of drug metabolic rate,mutation rate and angiogenic cells on drug resistance of patients and gave constructive treatment advice.In Chapter 3,we summarize the main research results obtained in this paper and put forward the deficiencies of this paper and future work prospects.
