BackgroundWhite adipose tissue reside underneath the reticular dermis has been shown to be a distinct adipose depot that modulate hair growth via secreting modulatory molecules.However,among those abundant diversity of adipose-derived cytokines,the major functioning molecules remain poorly defined.It is of great significance to clarify the role of adipose tissue in regulating hair follicle growth for clinical research on new treatment of hair loss disease.Therefore,we aim to thoroughly investigate the role of DPT in modulating hair growth.Objective1.To analyze the gene expression patterns of dermal white adipose tissue(dWAT)in different hair cycle stage,sort out and identify the dominating adipose-derived regulatory molecules via skin immunofluorescence,q-PCR,and WB techniques.2.To confirm the effect of dermatopontin on hair growth.Methods1.To analyze the gene expression patterns of dWAT in different hair cycle stage,sort out and identify the dominating adipose-derived regulatory molecules via skin immunofluorescence,q-PCR,and WB techniques.Firstly,RNA-seq sequencing was used to analyze the gene expression profiles of dermal white adipose tissue during different hair cycles.The expression pattern of dermatopontin during different hair cycles was examined by immunofluorescence and western blot.2.To confirm the effect of dermatopontin on hair growth.DPT protein was applied to examine the effect of DPT on in-vivo and in-vitro hair follicle growth.Results1.To analyze the gene expression patterns of DPT in different hair cycle stage,sort out the dominating adipose-derived regulatory molecules,and verify the expression pattern via skin immunofluorescence,q-PCR,and western blot techniques.The genes related to vascularization,hair follicle growth and development,extracellular matrix and melanin biosynthesis were significantly up-regulated during anagen.Immune system process related genes were up-regulated in catagen.Intelogen,DEGs mainly associated with negative regulation of biological process were significantly up-regulated.2.To confirm the effect of dermatopontin on hair growth.0.01ug/ml DPT protein inj ection on the back of mice can induce the transformation of hair follicle cycle from resting stage to growth stage.The total length of in-vitro cultured human hair follicle was significantly increased after DPT treatment.ConclusionDermal adipose tissue can promote hair follicle via up-regulating the modulatory molecules in anagen.In this experiment,we revealed the transcriptional characteristics of dWAT at different stages of the hair cycle and the important role of DPT derived from dWAT in regulating hair follicle growth.Our study may provide theoretical basis for new drugs invention to promote hair growth.
