Actinomycetes are widely used in various fields,especially in the medical field.Actinomycetes are an important source of antibiotics.The widespread use of antibiotics has led to the emergence of a large number of clinically resistant bacteria.Halophilic actinomycetes are a class of actinomycetes that are extremely tolerant to high-salt environments.They have a unique physiological mechanism to adapt to high-salt environments and may produce valuable secondary metabolites.Using high-throughput sequencing technology to study the actinomycetes resources in 28 samples of Aiding Lake,Dabancheng Saline Lake and Wuzunbulak in Xinjiang,the results showed that: Aiding Lake is mainly composed of Solirubrobacterales 15.5%,Gaiellales 13.2%,Niteiliruptorales 10.61% and Frankiales 10.05%;Dabancheng Saline Lake is mainly composed of Niteiliruptorales 23.04%,Actinomarinales 21.73%,Micrococcales 12.33%,Microtrichcales 11.70%;Uzunbulak is mainly composed of Euzebeyales 42.19%,Frankiales 18.48%,and Corynebacteriales 13.3% composition.The statistical results of classification information show that the coverage of the three salt lake samples to the actinomycete group is very high;the sample hierarchical cluster analysis shows that there are obvious differences between the three salt lake samples.In the process of pure culture separation,neomycin is used to eliminate the interference of halophilic archaea and most of the actinomyces polysporus are resistant to erythromycin and erythromycin analogues to establish the selective separation of antibiotics.method.The cultivable results show that the isolated actinomycetes include Saccharomonospora,Haloechinothrix,Halopolyspora,Thermobifida,and Thermobifida.Haloactinospora(Haloactinospora),in which 2 new candidate genera and 6 new candidate species were isolated from pure culture.The experimental results show that neomycin has a good inhibitory effect on archaea,and the probability of a new group of isolated actinomycetes is higher,which provides a new idea for the isolation method of salt lake actinomycetes.A new candidate genus YIM S02039 of halophilic actinomycetes was obtained by selective isolation of antibiotics,and a preliminary study on its multi-phase classification was preliminarily determined to be a new genus of Nocardiopsaceae.In this study,through high-throughput sequencing,we have gained an overall understanding of the composition and differences of the three salt lake communities in Xinjiang,and revealed the mining of the three salt lake actinomycetes resources.In the process of pure culture and isolation of three salt lake actinomycetes resources in Xinjiang,only considering the individual conditions of antibiotics,the probability of isolating new species will be greatly improved,which has guiding significance for pure culture and isolation of halophilic actinomycetes in other regions.
