The secondary metabolites of plant endophytic fungi are an important part of the secondary metabolites of microorganisms,and they have the characteristics of diverse types and rich biological activities.In this study,two endophytic fungi Trichoderma koningiopsis and Irpex lacteus obtained from Angelica sinensis were identified.The secondary metabolites and biological activities of above two endogenous fungi were investigated systematically.In chapters 1 and 2,the research background and target strain identification were described.Chapter 1,the medicinal plant Angelica sinensis is separated and purified different kinds of compounds that also briefly introduced the mechanism of biocontrol bacteria.Chapter 2 mainly used the observation of fungal morphology(growth time gradient)and molecular biology methods(neighboring optimization positioning method,etc.)to determine the target strains MK696749 and MT276332 as T.koningiopsis and I.lacteus.In chapter 3,it was carried out that systematic exploration and study of the chemical composition of T.koningiopsis solid fermentation,17 monomer compounds were isolated and identified,including 5 new compounds,konindines A-E of 6+6 bicyclic polyketone skeletons.The new compounds had the same basic skeleton,which some of the substituents were different or epimeric.When identifying the compounds konindines A-B,nuclear magnetic resonance data was used to determine the planar structure,and single crystal X-ray combined and ECD experimental data to determine the absolute configuration.When exploring the biological activity of monomer compounds,the main products were mainly analyzed biological activity.The inhibitory effect of the main products was 256?g/m L(MIC),and the NO production inhibitory activity of konindine D was 11.52±1.22(IC50?M).In chapter 4,it was carried out that systematic exploration and study of the chemical composition of I.lacteus solid fermentation,which 17 monomer compounds were isolated and identified.The main products of the secondary produced by the metabolism of this strain was 5+7 time the seaweed semiterpenes,with including two new compounds.The chapter 4also provided a brief overview of the biological metabolic pathways.When identifying new compounds,nuclear magnetic resonance and DP4+conformation optimization and error value analysis were used to identify new compounds.When exploring the biological activity,the main products were analyzed the antibacterial experiment and vitro anti-acetylcholinesterase activity.The E.coli inhibitory activity of Ceriponol A was 128?g/m L(MIC).This study shows that the endophytic fungi of Angelica sinensis has a rich variety of secondary metabolites,which can be used as fungal resources to isolate more complex and novel microbial secondary metabolites with medicinal value,thereby enriching the natural product resource library.
