Objective: The microarray data of gene expression profile of osteosarcoma were analyzed by bioinformatics analysis technology,so as to screen the hub genes related to the occurrence,development and prognosis of osteosarcoma,and reveal its potential molecular mechanism.Methods: Four data sets of GSE42352?GSE33382?GSE21257 and GSE39055 were obtained from GEO database,the differential expression of the first two data sets is analyzed,and the DEGs is intersected in the form of Venn diagram.The DEGs obtained from the intersection was analyzed by GO and KEGG enrichment analysis,and the PPI network was drawn to screen the hub genes.Then GSE21257 data sets was used to further verify the expression level of hub genes in metastasis group and non-metastasis group.Finally,the survival data of GSE39055 data set was used to analyze the relationship between hub genes and the prognosis of osteosarcoma patients.Results: A total of 386 DEGs,were obtained,including 204 up-regulated genes and 182 down-regulated genes.GO enrichment analysis showed that DEGs was mainly involved in biological processes such as immune response and negative regulation of RNA polymerase II promoter transcription,in terms of cellular composition,it mainly included extracellular matrix and cytoplasmic membrane,and molecular function involved protein binding and the same protein binding.KEGG pathway enrichment analysis showed that DEGs was mainly involved in cell adhesion molecules,osteoclast differentiation,proteoglycan,ECM receptor interaction and p53 signal pathway in cancer.Ten core genes were screened from PPI network,among which IGFBP4,GAS6 and IL6 showed a low expression trend in the occurrence and development of osteosarcoma.Prognostic analysis showed that the expression levels of GAS6,IGFBP3,CYR61,LAMC1 and FSTL3 were related to the prognosis of patients with osteosarcoma.Conclusion: In this study,hub genes IGFBP4,GAS6 and IL6,which are closely related to the occurrence and development of osteosarcoma,have been identified to increase people's understanding of the mechanism of occurrence and development of osteosarcoma.The hub genes GAS6,IGFBP3,CYR61,LAMC1 and FSTL3,related to the prognosis of osteosarcoma were screened,which provided new prognostic markers and potential therapeutic targets.
