Plant viruses are important pathogens that cause many serious plant diseases in the world and thus become threats for agricultural productions.It has been more than 100 years since the first plant virus was discovered.To date,there are more than 1,000 plant viruses have been identified.There is no effective chemical treatment to eradicate plant viruses,however,some cultural practices and biological control such as viral cross protection methods can be used to control the viral diseases and reduce plant yield losses.In this study,we demonstrated that asymptomatic infection of a DNA virus in Nicotiana benthamiana induces resistance or tolerance to infection of virulent heterologous RNA viruses based on cross protection mechanism.Apple geminivirus(AGV)is a geminivirus that infects apple trees in China.AGV does not cause symptoms in apple plants,therefore it is potentially to be used as an attenuated viral strain in viral cross-protection.In addition,AGV is a circular DNA virus,which is convenient for bioengineering technology.The main conclusions in this research:1?AGV infectious clone could systematically infect N.benthamiana for a long period.2 ? N.benthamiana that had been previously inoculated with AGV could resist the infection of PVX and CMV.3?AGV infectious clone could reduce the symptoms of PVY.4?AGV infectious clone could greatly delayed the time to death of N.benthamiana due to TMV infection.In this study,AGV infectious clone was constructed,and its infectivity in N.benthamiana was assessed.AGV infectious clone could systematically infect N.benthamiana for a long period.These results show that AGV is potential to be used for viral cross-protection against different types of plant virus diseases,and thus this study lay the foundation for further development of control methods of broad-spectrum of plant virus diseases using DNA viruses.
