Microorganisms are one of the important sources of active metabolites,which were developed to be as antibiotics,pest control drugs,tumor suppressors,immunosuppressants and so on.So far more than 20,000 active secondary metabolites of microorganisms have been found.In the continuous research on further exploring the active secondary metabolites from microorganisms,it was found that the content of target metabolites in microorganisms was low,which could not be enough for further research.It is a hot spot in the research of microbial natural products that the active secondary metabolites in microorganisms are explored through the screening of culture conditions and optimal separation techniques.The purpos of this dissertation is to search for secondary metabolites of active terpenes and peptides from Stereum hirsutum FP-91666 and Lysobacter gummosus03100.By screening the culture conditions,the optimal conditions were selected to further expand fermentation.The traditional separation technology and LC-MS guiding were used to find the microbial secondary metabolites.The structures of the compounds was determined by analysis of spectral data(NMR,MS,Optical Rotation and UV),and their activity was assayed.In the course of chemical investigation,based on the screening of the culture conditions of Stereum hirsutum FP-91666,the modification PWJ broth showed that Stereum hirsutum would produce more potential secondary metabolites.After fermentation,27 compounds were isolated and identified from the extracts of fermentation,7 of them were new compounds including one vibralactone type compound(1),two sesquiterpenoids(2 and 3),and three sesquiterpene-?-amino acid quaternary ammonium hybrids(4,5 and 6)and a new glycoside(7)as well as 20 known compounds including sesquiterpenoids,monoterpenoid,aromatics,furan rings,indoles and cyclodipeptides.The structures of 28 and 29 are still identifying.Five compounds were isolated from Lysobacter gummosus 03100 on SDA broth.Compounds 30,31 and 32 are Pyrrole class compounds,and compound 34 is cyclicdipeptide compound.The NMR spectra of compound 35 showed it would be a cyclic peptide,and the structure is elucidating.The antibacterial activity of new compounds was assayed,including Salmonella typhi and Staphylococcus aureus.The antibacterial activity was assayed by Microtitre Broth Dilution Method for initial concentration at 100 ?g/mL.The results showed that new sesiterpenoids had no obvious activity against Salmonella typhi and Staphylococcus aureus.
