Salivary gland tumors are mostly benign in histology,among which pleomorphic adenoma is the most common,accounting for 40%-45% of salivary gland benign tumors.Its biological behavior is between benign and malignant tumors,with the characteristics of borderline tumors,with potential malignant biological behavior,and its recurrence,invasion and even metastasis are often reported.Pleomorphic adenoma of salivary gland is often called mixed tumor because it contains mucoid tissue,chondroid tissue,neoplastic myoepithelial cells and other components.The development of tumor is regulated by many kinds of cytokines and signal pathways.Many scholars at home and abroad have found that the benign tumors of parotid gland are various in pathology.The benign tumors of submandibular gland are mainly pleomorphic adenomas,and there are great differences in the incidence rate of pleomorphic adenoma of two glands.There are many researches on the causes and pathogenesis of pleomorphic adenoma of salivary gland.But it is also salivary gland.Why is there a large difference in incidence rate of pleomorphic adenoma between parotid gland and submandibular gland? What is the cause of this phenomenon? There is no reasonable explanation at present.Bioinformatics is a science that uses computer as a tool to store,retrieve and analyze biological information in the research of life science.It is one of the important frontier fields of life science and natural science,and will also be one of the core fields of Natural Science in the twenty-first Century.Its research focuses on two aspects: genomics and proteomics.Specifically,starting from the sequence of nucleic acid and protein,it analyzes the biological information of structure and function expressed in the sequence.The research object of transcriptome sequencing is the sum of all RNA that a specific cell can transcribe under a certain functional state,mainly including m RNA and non coding RNA.Transcriptome research is the basis and starting point of gene function and structure research.Through the new generation of high-throughput sequencing,almost all transcriptome sequence information of a specific tissue or organ in a certain state can be obtained comprehensively and quickly.It has been widely used in basic research,clinical diagnosis,drug research and development and other fields.In this study,we used fresh samples of submandibular gland and parotid gland tumor to sequence the tissue expression profile,cluster analysis of biological functions and real-time real-time PCR(quantitative real-time PCR: q PCR)detection to explore the differential expression genes in the benign tumors of parotid gland and submandibular gland.To explore the pathogenesis,diagnosis,treatment and even prevention of pleomorphic adenoma of salivary gland.Objective: To screen out differentially expressed genes in benign tumors of parotid gland and submandibular gland and analyze their functions.This provides a new way of thinking for explaining the causes of the difference between the incidence rate of pleomorphic adenoma in benign tumors of parotid gland and submandibular gland.Methods: Fresh samples of parotid gland,submandibular gland benign tumor and surrounding normal gland were collected,total RNA was extracted by Trizol,m RNA was screened by poly(a)magnetic beads,c DNA library was constructed and sequenced by illuminate hi SEQ.The difference was statistically significant between P < 0.05 and FDR < 0.05.Using ingenuity pathway analysis(IPA)to analyze transcriptome characteristics,including signal pathways,upstream regulatory molecules and cytokines.The differential genes were detected by real-time fluorescence quantitative PCR.Results: It was found that WLS,Wnt11,LRP6,fzd2,fzd7,Dkk3,FZD1 and Dkk3 were the high expression genes in Wnt signaling pathway,and WLS expression in submandibular pleomorphic adenoma was significantly higher than that in parotid pleomorphic adenoma.Fzd10 expression was low in pleomorphic adenoma.MCC was highly expressed in tumors,but there was no significant difference between parotid gland and submandibular gland.LRP6,Wif1,fzd2 and fzd7 were low expression in worthinoma.From the results of go enrichment analysis,we found that the tissue and organ development of parotid gland and submandibular gland benign tumors in biological process,Sr dependent membrane targeted co translation protein,membrane targeted co translation protein,serine phosphorylation were significantly enriched,ribosome,PML main body,cytoplasm were significantly enriched in cell composition,ribosome composition,cytokine activity,guanosine acid interaction in molecular function The exchange factor complex and serine / threonine kinase activity were significantly enricheConclusion: There are some significant differences between the submandibular gland and benign parotid tumors.The WNT signaling pathway is different in these genes.It may provide a new direction for exploring the incidence rate of pleomorphic adenoma in the benign tumors of the submandibular gland and parotid gland.
