Escherichia coli is a conditional pathogen widely existing in nature.In recent years,the antibiotic resistance of farms has been increasing due to the abuse of antibiotics,especially the emergence of multidrug-resistant Escherichia coli,which has not only caused substantial economic losses to the breeding industry.At the same time,it also poses a considerable threat to public health security.The development of gene sequencing technology has made microbial whole-gene sequencing more convenient.With the reduction of sequencing costs and the improvement of sequencing precision,it provides a convenient tool for the study of bacterial resistance.The second-generation high-throughput sequencing technology represented by the Ion Torrent sequencing platform has the characteristics of simple operation,less time,and accurate results.Many types of research,such as genetic diseases,cancer,reproductive health,human identification and infectious diseases,have widely used this application.In particular,deep whole-genome sequencing of microorganisms has become the ultimate tool for discovering various genetic mutations such as SNPs,insertions,deletions,inversions,and complex recombination.It is suitable for identifying and discovering of bacteria and the typing and functional analysis of specific bacteria.The research of the bacterial resistance shows broad application prospects.This study used the OptiRead microbiological identification and drug sensitivity analysis system to perform a drug sensitivity test on multi-drug resistant E.coli JL05.The results showed that JL05 tested ?-lactam,cephalosporin,aminoglycoside,the antifolate,quinolone,macrolide,tetracycline,phenylpropanols,nitrofurans,and other 15 types of antibiotics are resistant to 15 kinds of antibiotics,only as a mediator of sulfisoxazole,sensitive to furantoin.After extracting the JL05 whole genome library complete construction,the whole gene was sequenced on the Ion Torrent sequencing platform.The sequencing data were preprocessed,spliced,compared,and assembled to obtain the whole gene map of JL05.Through the annotation and comparison of GO,KEGG,PATRIC,VFDB,CARD and other network databases,analyzing the gene characteristics of JL05.Based on the completion of gene function classification and analysis of metabolic pathways,72 virulence genes of 20 types in 6 categories were predicted.Power genes,67 types of drug resistance genes were found in 4 classes,which confirmed that the JL05 drug resistance phenotype and genotype are similar but also provided a scientific basis for further research on the JL05 drug resistance mechanism.
