The body cavity and water-vascular system of Apostichopus japonicus are filled with coelomocytes which are mainly responsible for nutrient transport and immune defense.Sea cucumbers have a special evisceration and regeneration mechanism.Some coelomocytes in the coelom are ejected with internal organs after evisceration,while coelomocytes in water-vascular system remain in the body.And the number of coelomocytes in the coelom increased rapidly at 6h post-evisceration.To further study the response of coelomocytes in coelomic cavity and polian vesicle of A.japonicus to evisceration and the role of polian vesicle in the recovery of coelomocytes,the variation of total coelomocyte count in polian vesicle(appendage of water-vascular system)of A.japonicus after evisceration was studied at first.Then the transcriptome analysis differences of coelomocytes between in coelomic cavity and polian vesicle were compared,and the transcriptome analysis of polian vesicle was also carried out.Transcriptome analysis showed that 267 genes of coelomocytes in polian vesicles were expressed differentially at 6h post-evisceration,while 922 genes of coelomocytes in coelom were expressed differentially.GO functional enrichment found that the differentially expressed genes of coelomocytes in polian vesicles were classified under in metalloendopeptidase inhibitor activity,peptidase inhibitor activity,and the differentially expressed genes of coelomocytes in coelom were classified under cell adhesion,biological adhesion.Analysis of differentially expressed genes through KEGG showed that these genes of coelomocytes in polian vesicles were significantly enriched in Glycine,serine and threonine metabolism pathway,and genes of coelomocytes in coelom were significantly enriched in ECM-receptor interaction,TGF-? signaling pathway and Fox O signaling pathway.Transcriptome analysis of polian vesicles in A.japonicus found that 2 752 genes of polian vesicle were expressed differentially.GO functional enrichment found that differentially expressed genes of polian vesicle were classified under regulation of macromolecule biosynthetic process,regulation of RNA biosynthetic process.Base on KEGG enrichment analysis showed that genes of polian vesicle were significantly enriched in TGF-? signaling pathway,Wnt signaling pathway and Fox O signaling pathway.The results of this study provide an important basis for further study on the function of coelomocytes and the mechanism of regeneration after evisceration.
