Dendritic spines are the structural basis for information exchange between neurons.Many factors regulate the density and morphological of dendritic spines,but the mechanism is unclear.There are three members in Disco-interacting protein 2(Dip2)family,Dip2a,b,and c.Bioinformatics analysis revealed that the Dip2 family has a highly conserved DMAP1 binding domain,AMP-binding domain and Cai C domain,which may be related to DNA methylation and lipid metabolism.To investigate the role of DIP2B in brain development,we prepared DIP2B antibodies.In this study,we found that DIP2B is widely expressed in the mouse brain and has a significantly higher expression in the cortex and hippocampus.At the same time,we found that the expression level of DIP2B increased gradually after birth,and the expression level began to increase significantly after 15 days.We constructed a Dip2b conditional knockout mouse,Dip2bflox/flox;Camk2-Cre mice,using the Cre-Loxp system to make DIP2B at specific sites which unable to express properly.Then we examined if Dip2b is knock out,there was any influence to dendritic in the cerebral cortex and hippocampal by Golgi staining.The results showed that the deletion of Dip2b resulted in the change of dendritic spine density in the hippocampal CA1,and the basal dendrites did not change significantly,but the apical dendrites showed a significant decrease.Further,through behavioral analysis,after the change of dendritic spine density,the open field experiment,the light and dark experiment,and the three-box interaction experiment did not show obvious abnormalities,but in the new object recognition experiment,the mouse which Dip2b was knockout can't find new object as wild type.In this study we showed that the expression of DIP2B in the mouse brain,and found that the deletion of Dip2b leads to the change of dendritic spine density in the CA1 region,and also have affect on behavioral,which provide an important theoretical basis for further study of Dip2b.
