Tumor immunotherapy has shown great potentials in cancer therapy.Specifically,adoptive cell therapy is in the process of rapid development.But in order to establish CAR-T cells,gene delivery is the key.Electroporation is an efficient and safe way of gene delivery,compared to viruses.But most devices use high voltages that may cause great damage to cells,and there may be toxic substances generate resulting from electrochemical reaction in buffer.All those defects limit the use of electroporation.Therefore,the purposes of our study were to build a cell electroporation device with minimum cell damage and high efficiency.We first,explore the use of nanostructures as electrodes.Gold-plated silicon nano-cylindrical array were made in the cell electroporation device.The transfection of adherent cells using nano-cylindrical electrodes was significantly better than transfection of the suspension cells.We then compared the effects using two kinds of electrode that were silicon nano-cylindrical electrodes and aluminum plate electrodes.The effect of suspended cells transfection with the plate electrode was better than silicon nano-cylindrical electrode,and damage of cells far less than commercial electroporation system.
