Cicadellidae belong to Hemiptera?Auchenorrhyncha?Membracoidea,which is the largest family of insects comprising more than 22,000 described species with worldwide distributions.But the relationship between the high-level system of Cicadellidae has been uncertain.Mitochondrial genomes have been used as an effective marker to study phylogeny of different taxonomic levels in the past decades with the rapid development of DNA sequencing and analytical methods,because of advantages such as the gene conservation,including gene rearrangements,indels ang mutations.To investigate the origin and diversification of Cicadellidae,we have sequenced the complete mitogenemes of 18 specimens of 13 subfamilies.Based on these datasets,the structure traits of Cicadellidae mitogenomes were analyzed using comparative genomic ang bioinformatics methods,otherwise,we reconstructed the phylogeny of Cicadellidae using complete mitogenomes.Conclusions were drawn as below:1.The size of the mitogenomes of Cicadellidae is between 148.kb and 16.7kb,there is no significant difference in the size of mitognomes between subfamilies.The nucleotide composition of the mitogenome is obviously AT bias,that means the use of AT is significantly higher than GC,and the use of AT is between 74.6% and 80.7%.2.Cicadellidae in the mitogenome,the arrangement of genes is relatively compact,most will exist between adjacent genes overlap phenomenon,the length of the overlap range from a few bases to dozens of bases;however,in the mitogenome of some species,the long non-coding region was found in the sequence(>200bp).3.The 37 genes in the mitogenomes of Cicadellidae are very conservative,and only 2 different permutations are found,and they are concentrated in the trnI-trnQ-trnM gene cluster.4.The secondary structure of rRNA of Cicadellidae is very similar to aphid,16S rRNA include five structural domains(??????? and ?),42 or 43 stem-loop structures;12 rRNA include three structure domains(??? and ?),24 or 26 stem-loop structures.5.The overall system development topology of this study is basically consistent.Deltocephalinae species alone together into one clade.Megophthalminae and Ulopinae,Iassinae and Coelidiinae,Macropsinae and Hylicinae showed the sister group relationship respectively.
