JUNCAO is a herb that has been screened through a three-level system screening method and has been scientifically proven to be suitable for cultivation of edible fungi and medicinal fungi.JUNCAO has not only economic functions but also ecological functions,social functions and cultural functions.In this experiment,Biolog technology,Illumina highthroughput sequencing technology and culture method were used to study the diversity of rhizosphere and endophytic microorganisms under different environmental condition.And antagonistic endophytes to plant pathogenic fungi were screened.The following results were obtained:Biolog technology was used to study the utilization of carbon sources by soil microorganisms.Analysis of AWCD values,six major sources of carbon sources,community diversity indices,and PCA principal components analysis showed that the microbial biomass of the JUNCAO in the rhizosphere soil has a high utilization rate of carbon sources,and the highest utilization rate is from the carbon sources of carboxylic acids,polymers,and amino acids..Based on the community diversity index,it was found that in the harsh environment of Inner Mongolia desert,rhizosphere soil microbial utilization of carbon sources was higher than non-rhizosphere soil.PCA principal component analysis shows that the degree of utilization of carbon sources in rhizosphere and non-rhizosphere varies greatly.Illumina high-throughput sequencing technology was used to analyze the rhizosphere and non-rhizosphere soil microorganism of the JUNCAO in the JUNCAO base of Fujian Agriculture and Forestry University Yangzhong Science and Education Center and JUNCAO base of Fujian Agriculture and Forestry University.16S and ITS sequencing indicated that the microbial richness of the soil and non-rhizosphere of the JUNCAO was significantly different.Among the 16S-sequenced samples,10 phyla were the most abundant,and Proteobacteria and Acidobacteria were the dominant bacteria in the samples.ITS sequenced samples had the highest abundance in 5 phyla,and Ascomycota was the predominant group in the samples.129 strains of endophytic fungi and 127 strains of endophytic bacteria were isolated from the roots,stems and leaves of the JUNCAO by using the method of plant tissue separation.They were identified to 12 species and 11 different genera,respectively.The antagonism of endophytic fungi and endophytic bacteria to Pyricularia oryzae by i was analyzed by plate-assisted method.A total of 69 strains of endophytic fungi were screened out.Among them,the strain JZRF3 had the highest inhibition rate of 70.5%.A total of 42 strains of endophytic bacteria were screened out,among which the inhibitory rate of the unknown strain NHG10 was the highest,and the inhibition rate was 80.3%.The control effect of endophytic bacteria from the JUNCAO against P.oryzae was analyzed by in vivo bioassay.The results showed that the fzg22 strain of Klebsiella pneumoniae had best control effect on P.oryzae.
