Establishment of 3D cell culture system for testing cellular responses to anti-cancer drugs

Abstract/Summary:
Monolayer 2D cell culture systems lack cues to fully understand how tissue form and function in microenvironments in vivo. 3D cell culture systems are currently being used to understand the signals that regulate function of tissues in normal and disease states such as cancer. Our study with Oral Squamous cell carcinoma cell line CAL27 includes establishing 3D model for CAL27 cell line, determining cellular responses in 3D culture to two drugs; Bleomycin and Erlotinib, compared to those of 2D and examining the expression levels of EGFR using western blotting.;Our results revealed that cells showed greater proliferation on undiluted Matrigel than in liquid media which is statistically significant. CAL 27 cells in 3D culture exhibited a more sensitive response towards both Bleomycin and Erlotinib treatments than in 2D culture. And the expression of EGFR protein was higher in cells growing on 3D compared to its expression in 2D.
