DNA sequence analysis of a Helicobacter pylori plasmid fragment

Abstract/Summary:
The purpose of this study is to determine the nucleotide sequence of a 700 bp plasmid DNA fragment isolated from the gastric pathogen, Helicobacter pylori. The DNA sequence has been analyzed for homology to other plasmid nucleic acid and amino acid sequence.; The 700 bp fragment was subjected to dideoxy chain termination DNA sequence and computer database analysis using DNAsis and PC-Gene program. One open reading frame was identified and the deduced polypeptide was 228 amino acids with predicted molecular weight of 27623. Three prokaryotic genes with short regions of DNA homology were found. However, no ribosome binding site and transcription termination sequences were revealed in the sequence. The 700 bp fragment must be part of an unknown protein. Sequencing the neighboring fragments may provide better identification of this plasmid-encoded protein.
