Transient replication assays and studies involving knockout viruses have identified seven proteins encoded by Autographa californica multiple nucleopolyhedrovirus that are essential for baculovirus DNA replication: IE-1, P143, LEF-1, LEF-2, LEF-3, DNAPOL, and LEF-11. However, the exact functions of most of these proteins have not been clearly established and little is known about how they assemble into a replication complex during viral infection. An existing protocol involving in vivo formaldehyde cross-linking and chromatin immunoprecipitations was modified for baculovirus study to identify the components of the viral replisome and to investigate the intracellular distribution and protein-DNA interactions of IE-1, P143, LEF-1, LEF-2, LEF-3, and DNAPOL in vivo.
