To determine thermal and rheological properties of emdex (dextrates monohydrates) using various lubricants

Abstract/Summary:
Dry granulation has been widely used in pharmaceutical industry as a tablet manufacturing process. Powders are basic materials used in almost all industries specially in tableting process. Different type of lubricants is use in tablet formulation which effect on property of raw material or other lubricants. Rheological and Thermal property of materials are useful tool for evaluate lubricant effect on other powder as well as characterizing a proper lubrication mixing time of lubrication powders. In this research we tried to develop a new method for evaluate lubrication effect on emdex and lubrication time using ft-4 rheometer, Effusivity sensor package (ESP), disintegration time, hardness of tablet and loss on drying measurements. Emdex, widely used excipients in pharmaceutical industry was granulated in Cuisinart mixer. In mixer 700 G Emdex were mixed with 1% different lubricants like magnesium stearate monohydrate (mg st-m), magnesium stearate dihydrate (mg st-d), Stear-o-wet (SOW) and mixture of mg st-m (94%) + SLS 9 (6%) to perform the time dependent study at different time interval. Specific energy (SE), Basic flow energy (BFE), Pressure drop, Bulk density and % Compressibility were measured using freeman technology powder rheometer (FT - 4). Samples were collected at each data point and effusivity measured at room temperature using ESP probe. Hardness, disintegration Time and loss on drying measurement tests were also done to determine proper lubrication mixing.;Emdex and 1% Stear-o-wet mixtures have good flow behaviors than other lubricants mixture due to the following reasons: Basic flow energy was drastically decreased than any other lubricant mixture. Specific energy was continues less than any other lubricant mixture. Pressure drop was also less than any other lubricant mixture. % Compressibility was very less than any other lubricant mixture. Effusivity data suggested dramatically increase in effusivity with increase in mixing time which show proper mixing between different lubricants. Hardness decrease dramatically with increase in mixing time also proves proper mixing with increase mixing time. Disintegration time follows same trend as effusivity which shows proper mixing. After that, better flow of granules was observed in presence of stear-o-wet. This observation made from the values of basic flow energy, specific energy, % Comressibility, pressure drop. Also proper mixing of lubrication mixture also observed with values of effusivity data, disintegration time and hardness data.;From the data we are able to quantify the changes occurring during dry granulation and determine proper lubrication mixture. The developed method is easy to perform, reliable and has less scale up issues.
