High yields of natural products in recombinant microorganisms using regulatory factors

Abstract/Summary:
Control of genetic expression, which is traditionally considered as protein's job, can also be achieved by small mRNA sequences located in 5'-UTR of any gene widely recognized as 'riboswitches'. These riboswitches control genetic expression by binding to a particular ligand which is typically a small metabolite or a metal ion. Here we explore the increase in expression of reporter gene lacZ by using theophylline binding riboswitch. At the same time control exercised by the promoter inducer arabinose is also tested through arabinose transporters regulation which is known to induce pBAD promoter system in an autocatalytic manner. This established system of dual expression control gives 10 fold increased gene expression as compared to wild type E. coli strain.;Ever increasing demand for effective drugs for diseases such as diabetes and other obesity related disorders have lead to exploring alternative options of natural products. Flavonoids which are one of the potential solutions for diabetes and obesity; are being used as case study to check the effectiveness of the already established system that uses riboswitches and controlled promoter induction for enhanced genetic expression.
