Abstract/Summary:
Implant-related bone infections are a major clinical and economic concern. Infections that are caused by bacterial adherence and colonization not only result in implant failure but also lead to the destruction of local tissues and sometimes even death. The conventional way to treat infection is through the use of systemic antibiotic regimens. It is increasingly difficult for systemic antibiotics to reach such a high concentration at local site without exceeding the cell toxicity level with increase in bacterial resistance towards antibiotics. This study showed the advantages of LbL (Layer-by-Layer) polyelectrolyte thin films as a platform for local delivery and controlled release of vancomycin on the surface of HAp (Hydroxyapatite) discs. In vitro studies demonstrated that vancomycin released from the LbL polyelectrolyte thin films on HAp surfaces significantly inhibited bacterial formation without having any significant affects on osteoblast viability. As such, LbL polyelectrolyte thin films may be used as a local drug delivery system with potential clinical applications for the prevention or treatment of orthopedic implant-related infections.
