As the improvement of human living quality and various life style,the morbidity of hypertension increases dramatically in nowadays and this situation causes a serious endangerments of people’s health.The artery vessel can be remodeled during the deteriorating process of the hypertension.These damages directly affect the function of human organ and kidney is one of most influenced organ,which will result in chronic kidney disease(CKD)when kidney experiencing long term of high blood pressure.Early clinical symptoms generate the micro-construction injured and lead to the epithelial-mesenchymal transformation.With the deterioration of the hypertension,it will induce the kidney fibrosis and decrease kidney function.Current studies discovered that the TGF-β1/Smad signal pathway is related to tissuefibrosis while decreased TGF-β 1expression of kidney could ameliorate kidney fibrosis.Moreover,exerciseis the most effective and safety method of treating.However,the exactly effect and the mechanism of exercise to hypertension kidney is unknown.Therefore,the aims of the present study was to examine whether exercise could ameliorate hypertension kidney injuredand if so,to determine the underlying mechanism,especially the role of TGF-β1/Smad signal pathway.Materials and method:SHR male rat and WKY male rats were used in the study.The animal were randomly assigned to three groups of 10 rats each as follows:group one,WKY,sedentary;group two,SHR,hypertension group;group three,SHRexercise group,Named SE group.After one week adapt training,SE group swam 60min a day within the second and the third week,thenswam 90min a day in the fourth,the fifth,the sixth and the seventh week.Finally,swam 120min a day in the last two weeks.After 10 weeks training,all the rats were euthanized,the blood and the samples were collected.Then abstracted the serum and preserved the renal in the-80 ℃ separately.Finally,executivethe histopathology test including HE staining,Masson staining,biochemical analysis and western blot.Result:1 Histopathology tests indicates that compared with the WKY rat,the glomeruli were swollen,the surrounding mesangium tissues also became thickened due to swelling,and extracellular mesenchyme were fibrosis in SHR,while the index of the SErat are all improved.2 The biochemical test(BUN/Scr)illustrate that renal function was decreasedinSHR including enhanced the amount of BUN and Scrin blood,while exercise improved the function bydecreasingthe amount of BUN and Scr.3 Western blotting test indicated that exercise down-regulated TGF-β1 Smad3 expressions and up-regulated Smad7 expressionin SHR,which exerted the benefit of exercise to the kidney fibrosis and renal function of hypertension.Conclusion:1 SHR exists injured renal with the fibrosis and decreasing function.2 Exerciseimproves renal function and renal fibrosis in theSHR at least partly via down-regulating the TGF-β1 Smad3 and up-regulating the Smad7expression of kidney.
