Objective:The problem of the juvenile crime is a serious social problem,especially violent crime,has received the widespread attention of the society.The juvenile crime is mainly through aggression to implement.In addition,violence is also frequent in the field of mental illness and sports field.The management of the attack has become a common concern of society.The research of aggressive behavior is not only can promote the development of basic science,but also have important practical significance for the improvement of human survival environment,the improvement of life quality.Research shows that central to the low level of 5-HT is associated with high attack.5-HT1AR and 5-HT1BR adjust the release of the 5-HT.Electrical stimulation of the medial hypothalamus cat induced defensive attack,found in the periaqueduetal grey 5-HT1AR plays an inhibition in attack.5-HT1BR knockout mouse performance is more sensitive to the unpredictable stimulation,are more likely to generate impulse.Our laboratory experiments have confirmed that sport can effectively improve the rat attack mood and behavior,but whether the different influence of different intensity exercise on aggression?It's not sure.Therefore,this study from the perspective of the exercise intensity,Choose the 5-HT,5-HT1AR,5-HT1BR and its mRNA expression in the 5-HT system of PAG area,the preliminary discussion on the influence of different intensity of swimming exercise on aggression.Methods:1.Animals and model60 adult healthy male Sprague-Dawley(SD)rats.Adaptive breeding a week later,randomly assigned:(A)Control group 9,(B)Attack model group 9,(C)Small intensity swimming model group 11,(D)Moderate intensity swimming model group 11,(E)Heavy intensity swimming model group 11,(F)Invasion group 9.(A)Control group:do not make any processing;(B)Attack model group:Single cage + invasion,3 weeks;(C)Small intensity swimming model group;single cage+ invasion,3 weeks;small intensity swimming training 8 weeks;(D)Moderate intensity swimming model group:single cage + invasion,3 weeks;moderate intensity swimming training 8 weeks;(E)Heavy intensity swimming model group:single cage + invasion,3 weeks;heavy intensity swimming training 8 weeks;(F)Invasion groups:invasion of single cage rat.2.Index and Method(1)Behavior:body weight,attack detection,open field test;(2)5-HT system of periaqueductal gray:Elisa:to test the serotonin(5-hydroxytryptamine,5-HT)in PAG;IHC:to test the expression of 5-HT1AR(5-HT1Areceptor),5-HT1BR(5-HT1B receptor)in PAG;RT-PCR:to test the expression of 5-HT1ARmRNA,5-HT1BRmRNA in PAG;Results:1.results show that against stress and exercise to lower body weight of rats in different degree;2.The attack detection found that,compared with A group,D group is presented significantly lower after 3 weeks exercise(P<0.05),and continues to the end of the training;E group is significantly lower after 3 and 5 weeks exercise(P<0.05),after 8 weeks is not significant;And compared itself,C group attack times extremely significantly lower(P<0.01)after 8 weeks exercise,D group attack times extremely significantly lower after the third time test(P<0.01),and continue to 12 weeks;E group significantly lower after the fourth test(P<0.05),and after the fifth test appears extremely significantly lower(P<0.01).3.Before modeling,each group of rats in the number of upright,modified frequency,total journey there was no significant difference.After modeling,compared with A group,the number of rats upright of B,C,D and E group were significantly increased(P<0.05),modified frequency significantly decreased(P<0.05),total journey significantly increased(P<0.05).After 8 weeks exercise,compared with A group,the number of upright of B group significantly increased,modified frequency extremely significantly lower(P<0.01),total journey significantly increased(P<0.05),between the C,D and E groups have differences,but not significant.compared with B group,C group modified frequency significantly increased(P<0.05);D group the number of upright extremely significantly lower(P<0.01),modified frequency significantly increased(P<0.05),total journey extremely significantly lower(P<0.01);E group modified frequency significantly increased(P<0.05),total journey significantly lower(P<0.05).Shows that Moderate intensity and Heavy intensity swimming had a greater influence on the aggressive rats in open field tests,and Moderate intensity swimming more significantly.4.B group 5-HT extremely significantly reduced by 27.8%in PAG(P<0.01).After 8 weeks swimming,compared with B group,D group extremely significantly increased by 44.37%(P<0.01),C group increased by 26.87%,E group increased by 21.99%,but not significant.5.B group 5-HT1AR expression extremely significantly reduced by 28.64%in PAG(P<0.01).C group significantly reduced by 20.31%(P<0.05).Compared with B group,C group increased by 11.68%,C and D group extremely significantly increased by 32.45%and 31.73%(P<0.001)?6.B group 5-HT1BR expression significantly reduced by 19.31%in PAG(P<0.05),After 8 weeks swimming,compared with B group,C group increased by 8.73%,D group significantly increased by 21.78%(P<0.05),D group increased by 12.44%.7.B group 5-HT1 ARmRNA expression significantly reduced by 21.8%in PAG(P<0.05).After 8 weeks swimming,compared with B group,C group increased by 9.6%,D group extremely significantly increased by 35.9%(P<0.01),E group increased by 18.3%.8.B group 5-HT1BRmRNA expression significantly reduced by 32.7%in PAG(P<0.01).After 8 weeks swimming,compared with B group,C group increased by 18.9%,D group extremely significantly increased by 47.3%(P<0.01),E group increased by20.4%.Conclusion:1.Single cage and invasion model can make the attack behavior in rats,the bite and fight,etc obviously increase and the exploration and vigilance increased in a novel environment;2.Aggressive rats' 5-HT below the normal rats,5-HT1AR?5-HT1BR and their mRNA expression both reduced;3.After 8 weeks swimming,the behavior of each swimming model group is improved,but the moderate intensity swimming more significantly.4.After 8 weeks swimming,moderate intensity swimming cause the 5-HT increased,5-HT1AR?5-HT1BR and their mRNA expression both raised;5.According to the study,aggressive rats'5-HT system reduced,Swimming can make the system function recovered to varying degrees,but the moderate intensity swimming more significantly.
