The incidence and mortality of primary liver cancer(hereinafter referred toas liver cancer)ranks fourth and second in my country's cancer morbidity andmortality,which seriously threatens the lives and health of our people.The current clinical treatment methods for liver cancer include hepatectomy,liver transplantation,radiochemotherapy,etc.,but the recurrence rate after treatment is still as high as 80%to 90%,and the quality of life of patients has not been effectively improved.The occurrence and development of cancer is a complex process involving multiple genes,multiple steps,multiple stages,and interactions between internal and external factors.Previous cancer therapies mainly targeted a single target,and cancer cells tend to develop resistance through compensatory pathways,leading to treatment failure.Multi-target therapy can act on multiple targets at the same time,enhance the efficacy,and it is not easy to make cancer cells resistant.Angiogenesis and apoptosis are critical to the development of cancer.The zebrafish has the characteristics of simple feeding,low cost,strongfertility,embryonic transparency,small individuals,rich genetic information,optical transparency,and a zebrafish tumor transplantation model based on the genetically modified zebrafish,which allows us to be more intuitive.The study of tumor cell growth in zebrafish and tumor-induced neovascularization in vivoDue to the multi-target mechanism of action of traditional Chinese medicin-e,in recent years,in the prevention and treatment of tumor diseases,traditionalChinese medicine has achieved good results in reducing the adverse effects of chemotherapy,improving the quality of life of patients and preventing the recurrence or metastasis of tumor cells.So that it shows attractive application prospects in the development of cancer drugs.Cinnamon is a traditional medicine and food homologous plant in China.Traditional Chinese medicine is commonly used in the treatment of various dis-eases.Modern pharmacological studies have shown that cinnamon can significantly inhibit cervical cancer,lung cancer,gastric adenocarcinoma,breast cancer,liver cancer and other cancer cells.proliferation.However,there are few studies on the effect of cinnamon on liver cancer.In the process of clinical treatment of tumors in traditional Chinese medicine,cinnamon is often used in the form of water decoction,so this topic takes cinnamon water extract as the research object to explore its efficacy and mechanism of inhibiting liver cancer.Early laboratory studies have found that cinnamon water extract has an inhibitory effect on angiogenesis in juvenile zebrafish.Combined with the literature,we speculate that cinnamon may inhibit tumor growth by promoting apoptosis and inhibiting the formation of neovascularization.In this study,human liver cancer Be17402 cells and tumor-bearing zebrafish models will be used as the research object.By measuring the cell proliferation inhibition rate,observing the cell cycle changes,determining the cell apoptosis and calculating the apoptosis rate,observing the tumor growth in vivo and observing the tumor induction.The growth of new blood vessels,study the effect of cinnamon water extract on the growth of liver cancer cells and explore its mechanism of actionThe experimental research is divided into three chapters,briefly described as follows:Chapter 1:After in vitro treatment of liver cancer Be17402 cells with different concentrations of cinnamon water extracts(50,100,200,400,800,1000 ?g/mL)for different times(24 h,48 h,72 h),MTT method was used to detect cell proliferation In the circumstances,it was found that with the increase of drug concentration and the prolonged action time,the proliferation inhibition rate of liver cancer Be17402 cells also increased significantly in sequence(P<0.01).Based on the MTT experiment results,cinnamon water extracts(200,400,800 ?g/mL)were seleced to interfere with liver cancer Be17402 cells for 48 hours.After the action was completed,the treated cells were collected and the cell cycle changes and Annexiv were detected by PI staining-flow cytometry./PI double staining method-flow cytometry was used to observe apoptosis and calculate the apoptosis rate.The experimental results showed that after intervention with cinnamon water extract for 48h,the proportion of human liver cancer Be17402 cells in G1 phase was significantly increased and in S phase The proportion of serotonin was significantly decreased(P<0.001);the intervention of cinnamon water extract(200,400,800?g/mL)for 48h could induce dose-dependent apoptosis of human hepatocellular carcinoma Be17402 cells,and the apoptosis rates were(16.49± 1.88,29.76 ± 1.80,53.27 ± 2.28).The above experimental results revealed that cinnamon water extract can inhibit the proliferation of human hepatoma Be17402 cells and promote the apoptosis of Be17402 cells,which may be related to its ability to block the cell cycle of Be17402 and regulate the expression of apoptosis proteins.Chapter 2:In order to study the changes of tumor blood vessels in vivo,this chapter uses transgenic zebrafish as a model,uses DiO dye to stain cells,and uses microinjection technology to construct a tumor-bearing zebrafish model.It provides a basis for the subsequent observation of tumor growth in vivo and tumor-induced growth of subintestinal blood vesselsChapter 3:Using tumor-bearing transgenic zebrafish as an animal model to detect the growth of tumor and tumor-induced subintestinal blood vessels in tumor-bearing transgenic zebrafish after different concentrations of cinnamon water extract(200,400,800 ?g/mL).The experimental results show that the 200-800 ?g/mL cinnamon water extract solution can inhibit the growth of tumors and the intestinal blood vessels of tumor-bearing zebrafish in a concentration-dependent manner.This study shows that cinnamon water extract has an inhibitory effect on liver cancer cells in vivo and in vitro,which may be related to the cinnamon water extract can block the cell cycle,promote apoptosis,and inhibit tumor neovascularization.This study provides a theoretical basis for the research of cinnamon in the treatment of liver cancer.
