With the rapid development of the economy and the accelerating pace of life,the prevalence and death rate of the psychocardiacology disease are increasing year by year,so it is crucial to find an effective treatment strategy.Our team's previous studies have shown that Chaihujialonggumuli soup can effectively improve the clinical angina symptoms and TCM syndromes of patients with coronary heart disease complicated with anxiety,reduce the anxiety state of patients,and improve the quality of life of patients without obvious adverse reactions,but the mechanism of action is not completely clear.The inflammatory response mediated by NF-?B was found to be a key factor in the development of coronary heart disease.NF-?B is active in neurons and is involved in the development,growth and apoptosis of nerve cells,playing an important role in the anxiety state.On the basis of the above background,this paper took myocardial infarction combined with anxiety as the representative of the psychocardiacology disease,and discussed the partial mechanism of Chaihujialonggumuli soup in the treatment of the psychocardiacology disease from the inflammatory response mediated by NF-?B.Objective:This paper discussed the inflammatory mechanism of Chaihujialonggumuli soup in the treatment of myocardial infarction complicated with anxiety in mice,providing a theoretical basis for the clinical treatment of the psychocardiacology diseaseMethods:The myocardial infarction combined with anxiety model was established in C57BL/6J mice by anterior descending coronary artery ligation combined with uncertain empty bottle stimulation.The successful mice were randomly divided into model group,sham operation group and Chinese medicine group(in Chaihujialonggumuli soup granules intervention),8 rats in each group were intragastrically administered daily.After 7 days of continuous administration,the cardiac function of the mice was evaluated by echocardiography,the pathological changes of the hearts were observed by myocardial Hematoxylin-eosin staining,the degree of myocardial fibrosis was observed by myocardial Masson staining,and the activity ability and anxiety state of the mice was evaluated by the open field test and the elevated plus maze.ELISA was used to detect the contents of IL-1?,TNF-? and IL-6 in peripheral blood.RT-PCR was used to detect the expression levels of IL-1?,TNF-? and NF-?Bp65 mRNA in the marginal area of myocardial infarction,and WB was used to detect the protein expression levels of NF-KBp65 in the marginal area of myocardial infarction.Results:1.Structural and functional aspects of the heartEchocardiography:compared with the sham group,the left ventricular ejection fraction and short axis shortening rate of the model group were significantly reduced,and the left ventricular end-diastolic volume and end-systolic volume were significantly enlarged(P<0.01).Compared with the model group,the left ventricular ejection fraction and short axis shortening rate of mice in the traditional Chinese medicine group were significantly increased,and the left ventricular end-diastolic volume and end-systolic volume were significantly reduced(P<0.01)HE staining:in the sham operation group,the myocardial cell structure was basically normal,the cytoplasm was uniformly stained,the nuclei were clearly stained,and the muscle fibers were neatly arranged.In the model group,there was a large amount of myocardial cell necrosis,nuclear fragmentation and myocardial muscle fiber fracture.The structure damage of myocardial cells and fiber fracture were alleviated in the Chinese medicine groupMasson staining:the myocardial cells in the sham operation group were arranged neatly,and no obvious collagen fibers were observed.In the model group,the arrangement of cardiomyocytes was obviously disordered,and the interstitium showed significant collagen fiber and inflammatory infiltration.In the Chinese medicine group,the myocardial cell arrangement disorder was reduced,the collagen fiber was reduced,and the degree of myocardial fibrosis was reduced.2.Anxiety stateThe open field test:compared with the sham group,the horizontal and vertical movement scores of mice in the model group decreased(P<0.01).Compared with the model group,the horizontal movement score and the vertical movement score of the mice in the traditional Chinese medicine group increased(P<0.01)The elevated plus maze test:compared with the sham group,the percentage of times the model group entered the open arm and the percentage of time the model group stayed in the open arm decreased(P<0.01).Compared with the model group,the percentage of the times and the percentage of the stay time of the open arm in the traditional Chinese medicine group increased(P<0.01).3.NF-?B mediated inflammationELISA method was used to detect inflammatory factors in peripheral blood.Compared with the sham group,the expression levels of IL-1?,IL-6 and TNF-? in the model group were increased(P<0.01).Compared with the model group,the expression levels of IL-1?,IL-6 and TNF-? in the Chinese medicine group were reduced(P<0.01).RT-PCR detection of mRNA expression levels of inflammatory factors at the edge of myocardial infarction:compared with the sham group,the expression levels of IL-1?,TNF-?and NF-?Bp65mRNA in the model group were increased(P<0.01).Compared with the model group,the expression levels of IL-1?,TNF-? and NF-?Bp65mRNA in the Chinese medicine group were reduced(P<0.01).WB detection of NF-?Bp65 protein expression level in the marginal area of myocardial infarction:compared with the sham group,the expression level of NF-?Bp65 protein in the model group was increased(P<0.01).Compared with the model group,the expression level of NF-?Bp65 protein in myocardial tissue of mice in the traditional Chinese medicine group was decreased(P<0.01).Conclusion1.Chaihujialonggumuli soup can improve cardiac function and the pathological changes in mice with myocardial infarction combined with anxiety;and reduce the degree of myocardial fibrosis;2.Chaihujialonggumuli soup could improve the anxiety-like behavior of the mice with myocardial infarction combined with anxiety;3.Chaihujialonggumuli soup can reduce the degree of inflammatory response mediated by NF-?B in mice with myocardial infarction combined with anxiety;4.Myocardial infarction may promote anxiety in mice by exacerbating the inflammatory response mediated by NF-?B;5.Chaihujialonggumuli soup may improve cardiac function and anxiety in mice with myocardial infarction by regulating the inflammatory response mediated by NF-?B.
