Background:Prostate cancer(PCa)is one of the most common malignant tumors in elderly men.The incidence rate of PCa is the highest in male cancer in Europe and the United States.The incidence rate of PCa in China is also increasing year by year.For localized prostate cancer,radical surgery or radiotherapy is the most effective treatment.When the tumor enters the late stage and cannot undergo radical treatment or postoperative recurrence,or distant metastasis,androgen deprivation therapy(ADT)is the main treatment,and most patients can get better to a certain extent.However,after a median time of 18-24 months,almost all patients will be converted to castration resistant prostate cancer(CRPC).The treatment of CRPC is recognized as a difficulty and hot spot in the world.Traditional Chinese medicine has a long history in the treatment of tumor,with its unique syndrome differentiation and treatment and holistic view as its advantages.Extracting and screening effective components from Chinese herbal medicine in the treatment of castration-resistant prostate cancer is a research direction that researchers at home and abroad pay more and more attention to.Geraniin is a kind of polyphenol compound widely distributed in nature,which has many biological activities,such as anti-oxidation,anti-tumor,anti-virus and anti-inflammatory.It has been found that geraniin can induce apoptosis,and inhibit proliferation,migration and invasion of lung cancer and melanoma cells by regulating cell-related signal pathways.In recent years,geraniin has been reported to have antitumor,antioxidant and antiviral properties.The purpose of this study is to explore the effect of geraniin on the proliferation and migration of castration resistant prostate cancer cells,to lay the experimental foundation for further study on the biological function of geraniin,to find the natural active compounds with potential clinical application value,and to provide more theoretical basis for the treatment of castration resistant prostate cancer by Chinese herbal medicine.Objective:To explore the effect of geraniin on the proliferation and migration of castration resistant prostate cancer cells,in order to provide more preliminary basis and ideas for further mechanism research.Method:In this experiment,human prostate cancer DU 145 cells were cultured,and the experimental concentration gradient and treatment time gradient of geraniin were selected.The cells were divided into two groups:control group and experimental group.The effect of geraniin on the proliferation of human prostate cancer cell line DU 145 was detected by MTS test,the migration ability of human prostate cancer cell line DU 145 was detected by scratch test and Transwell test,and the proliferation and migration related protein expression of human prostate cancer cell line DU 145 was detected by Western blot test.Results:The results of MTS showed that geraniin significantly inhibited the proliferation of human prostate cancer DU 145 cells in a concentration dependent manner(P<0.05);scratch test and Transwell test showed that geraniin significantly inhibited the migration of human prostate cancer DU 145 cells(P<0.05);after geraniin intervention,western The expression level of c-myc protein was significantly lower than that of the control group,and the expression level of E-cadherin protein was significantly higher than that of the control group.Conclusion:Geraniin can inhibit the proliferation of DU 145 cells by down regulating the expression of c-myc and affect the epithelial-mesenchymal transformation of DU 145 cells by up-regulating the expression of E-cadherin,thus weakening its cell migration ability.Geraniin can significantly inhibit the proliferation and migration of castration resistant prostate cancer cell line DU145.
