OBJECTIVEObserve the effects of acupuncture of "Dazhui","Shenshu" and "Zusanli" on weight loss,neurological deficit score,brain tissue demyelination,and brain tissue in experimental autoimmune encephalomyelitis(EAE)mice The effects of phosphorylated P38 mitogen-activated protease and IL-17 to verify the therapeutic effect of acupuncture on EAE,explore the relevant mechanisms of acupuncture in the treatment of EAE,and provide a theoretical basis for the clinical treatment of multiple sclerosis(MS).METHODSForty female C57BL/6 mice were randomly divided into a blank group,a model group,a hormone group,and an acupuncture group.The mice in the blank group were not treated,and the remaining groups used myelin oligodendrocyte glycoprotein 35-55(MOG35-55)The prepared complete antigen induces mice into the EAE model.Starting on the 14th day after modeling,the blank group and model group were fixed daily,and intragastrically administered with normal saline.The hormone group was given intragastrically daily with prednisone acetate.Acupuncture group mice acupuncture "Dazhui","Shenshu" on both sides and "Zusanli" on both sides.Record the body weight and nerve function score of the mice.On the 28th day after immunization,brain tissues were collected and stained with LFB to observe pathological changes.Immunohistochemical expression of p-p38MAPK and WB expression of p-p38MAPK in brain tissue were detected,and IL-17 expression level in brain tissue was detected.RESULTS(1)Changes in mouse body weight:At the beginning of the experiment,the weight of the four groups of mice was roughly the same(P>0.05);on the 10th day of immunization,the weight of the mice in the blank group was greater than that of the model group,acupuncture group,and hormone group(P<0.05).There was no difference in the body weight of the mice in the acupuncture group and the hormone group(P>0.05);after the 22nd day of immunization,the mice in the model group weighed less than the acupuncture group and the hormone group(P<0.05),and the mice in the hormone group weighed more than the needle Acute group(P<0.05).(2)Mouse neurological deficit score:On the 10th day of immunization,there was no difference in the neurological deficit scores of the model group,acupuncture group and hormone group(P>0.05);on the 16th day of immunization,the scores of the acupuncture group and hormone group were lower than the model group(P<0.05),there was no difference in score between the acupuncture group and the hormone group(P>0.05);after the 22nd day of immunization,the score of the acupuncture group was higher than that of the hormone group(P<0.05).(3)Staining of myelin in LFB of brain tissue:the brain cells in the blank group had a normal structure,the cells were tightly packed,and the myelin sheath was dense without loss;the brain cells in the model group were loosely arranged,and a large number of demyelination cells were seen,showing vacuolar-like changes;In the acupuncture group and hormone group,some brain cells were demyelinated and scattered vacuoles were found,but the demyelination sites were less than the model group,and the degree was lower than the model group.(4)Immunohistochemistry and Western blot detection of brain tissue p-p38MAPK:the blank group had the lowest p-p38MAPK protein expression level,and the model group had the highest p-p38MAPK protein expression level.The difference between the two was significant(P<0.05).The p-p38MAPK protein expression level in the hormone group was higher than that in the blank group(P<0.05),but lower than the model group(P<0.05).There was no difference in the p-p38MAPK protein expression level between the acupuncture group and the hormone group(P>0.05).(5)IL-17:the blank group had the least IL-17 content in the brain tissue of the mouse,the model group had the most IL-17 content in the brain tissue of the mouse,and the difference in the IL-17 content in the brain tissue of the two groups was statistically significant(P<0.05);IL-17 content in brain tissue of acupuncture group and hormone group was higher than that of blank group(P<0.05),but lower than model group(P<0.05),IL of acupuncture group and hormone group mice There was no difference in-17 content(P>0.05).CONCLUSIONS(1)Acupuncture can reduce the weight loss of EAE mice and lower the neurological score of EAE mice.(2)Acupuncture can reduce the degree of demyelination in the brain of EAE mice,suggesting that acupuncture can inhibit the demyelination of brain tissue.(3)Acupuncture reduce the expression of p-p38MAPK in the brain of EAE mice,suggesting that acupuncture can inhibit the p38MAPK signaling pathway.(4)Acupuncture can reduce the expression of IL-17 in the brain of EAE mice,suggesting that acupuncture can reduce the inflammatory response.
