Endoscopic Optical Coherence Tomography(EOCT)is a combination of optical coherence tomography(OCT)and endoscopic techniques.It has the advantages of fast imaging speed and high resolution,and can be used in imaging of live organs in vivo and real-time imaging.EOCT has become one of the focused areas of bioimaging technology.In this thesis,the design and construction of an EOCT system which was based on the common path endoscopic probe and was suitable for human esophageal detection was investigated.First of all,the principle of imaging OCT system in the frequency domain was studied,and the system composition and imaging system parameters were analyzed,with the emphasis on the design methods which are the most critical in the design of the endoscopic OCT system.Second,a design of the common path endoscopic probe based on fiber-glass-ferrule was proposed.The fiber-glass-ferrule integrated the detecting arm and the reference arm into the endoscopic probe,which can increase the stability and the ease of debugging of the system;Third,the Gaussian optical ABCD transmission matrix was utilized to establish the mathematical relationship between the spot size,the working distance and the air gap between the fiber and the GRIN lens.The air gap length was determined on the basis of suitable for the esophageal detection.Fourth,the manufacturing processes of the endoscopic probe was established,and the manufacturing of the internal optical components of the probe as well as the assembly of the probe was completed.Finally,the construction and debugging of the entire endoscopic OCT system was completed,and the performance parameters of the system were analyzed.By using a high-speed industrial camera,the spot size and working distance of the endoscopic probe was verified.The work described in this thesis provides the basis for future applications of the endoscopic OCT system for in vivo imaging of internal structures of human organs.
