Dendrobium huoshanense is one of the most prized medicinal herbs in China with a long medicinal history.There are a variety of pharmacological activities of Dendrobium huoshanense such as anti-inflammatory,anti-oxidation,improving human immunity,anti-cataract and liver protection.Dendrobium huoshanense contains several main components including polysaccharides,alkaloids,flavonoids and trace elements.In recent years,wild Dendrobium huoshanense has been overexploited and endangered.So most of Dendrobium huoshanense in the market is planted artificially.About the harvest time of Dendrobium huoshanense,most of the harvest months are from October to June every year,but there is no relevant literature report about the specific harvest year of Dendrobium huoshanense.However,the content of effective components of different growth years of medicinal materials is quite different,which will directly affect the clinical efficacy of medicinal materials.Therefore,the research on the effective components and efficacy of different growth years of Dendrobium huoshanense would provide a basis for the development of Dendrobium huoshanense.In this study,the contents of polysaccharides,total flavonoids,total alkaloids and gigantol of Dendrobium huoshanense of different growth years were determined.The differences of the efficacy of liver protective were studied by constructing the model of liver injury of mice induced by paracetamol?APAP?.Analysis of the relationship between the components and the efficacy of Dendrobium huoshanense of different growth years was conducted by SPSS.In addition,the anti-inflammatory activity of Dendrobium huoshanense of different growth years were studied.This study provides a basis for the reasonable harvest year of Dendrobium huoshanense and the industrial development of Dendrobium huoshanense.The main research contents and conclusions are as follows:1.The differences of main components of Dendrobium huoshanense of different growth years.The content of polysaccharide,total flavonoids and total alkaloids of Dendrobium huoshanense of different growth years were determined by UV spectrophotometry,and the content of gigantol of Dendrobium huoshanense in different growth years was determined by HPLC.The results showed that the chemical components of Dendrobium huoshanense of different growth years were different.The polysaccharide content of Dendrobium huoshanense of two growth years was the highest,which was 159.34 mg/g,and that of Dendrobium huoshanense of one growth year and three growth years were 141.17 mg·g-1 and 101.23 mg·g-1,respectively.The content of gigantol of Dendrobium huoshanense of two growth years was also the highest,reaching 12.58 mg·g-1,that of Dendrobium huoshanense of one growth year and three growth years were 5.93 mg·g-1 and 4.60 mg·g-1,respectively.The contents of flavonoids and alkaloids of Dendrobium huoshanense of three growth years reached7.41 mg·g-1 and 0.58 mg·g-1 respectively and were the highest,followed by the Dendrobium huoshanense of two growth years reaching 4.91 mg·g-1 and 0.48 mg·g-1,and the lowest were that of one growth year reaching 3.28 mg·g-1 and 0.09 mg·g-1,respectively.2.The protective effects of Dendrobium huoshanense of different growth years on acute liver injury in mice induced by acetaminophen.Acetaminophen is a common antipyretic,analgesic and anti-inflammatory drug,but large dose of drugs or long-term use will lead to liver injury.In this study,acetaminophen liver injury model was used to evaluate the efficacy of Dendrobium huoshanense of different growth years.The results showed that all administration groups could decrease the activities of ALT and AST in serum.Meanwhile,the levels of MDA reduced significantly,while those of SOD increased markedly.Histopathological results suggested that all Dendrobium huoshanense samples were effective in the reduction of the necrosis of hepatocytes in different degrees.But the efficacy of Dendrobium huoshanense of different growth years were different,and the efficacy of two growth years was the best.The results of the analysis of SPSS showed that polysaccharide and gigantol probably played a leading role in the liver protection effects.3.The anti-inflammatory effects of Dendrobium huoshanense of different growth years.The anti-inflammatory effect of Dendrobium huoshanense of different growth years was evaluated by ear swelling model in mice.The results showed that Dendrobium huoshanense of different growth years could reduce the ear swelling of mice in different degree and Dendrobium huoshanense of two and three growth years showed the best efficacy.
