Objective: PAEs is the most widely used plasticizer,which can be added to PVC products to give flexibility and durability.It is widely used in building materials,food packaging,blood bags,intravenous infusion bags,infusion sets and other medical equipment,but at the same time,phthalate esters also have great harm to people's health.DINP,as a new plasticizer,has the advantages of high boiling point,low melting point and low toxicity.It is the most promising environmental plasticizer in PAEs.A number of studies have shown that DINP exposure may be associated with the development of allergic asthma.The adjuvant effect of DINP on allergic asthma may be the way to promote immune response,but these findings are still speculative,because there is not enough evidence to evaluate the ability of DINP to affect the "pathology of allergic asthma".The purpose of this study is to determine the effect of DINP exposure on all aspects of the airway of the allergic asthma model by constructing a mouse model of allergic asthma.We hope our study can provide a scientific basis for clarifying the molecular mechanism of DINP induced allergic asthma.Methods: This study was to investigate whether DINP has adjuvant effect on allergic asthma.In the experiment,we used ovalbumin(OVA)and aluminum hydroxide colloid to sensitize,and activated with 1% ova solution.Airway hyperresponsiveness(pulmonary function),lung histopathology(H&E,Masson and PAS),Th1/Th2 cytokines(IFN-?,T-IgE,OVA-IgE,IL-5,IL-13 and IL-33)and oxidative stress biomarkers(ROS,GSH,MDA,NO)were detected.To study whether DINP has adjuvant effect on allergic asthma.Results: The results showed that the level of Th1 and Th2 cytokines in ova group was significantly lower than that in normal saline group,which resulted in the imbalance of Th1/Th2 immunity;airway hyperresponsiveness and lung histopathological damage were significantly aggravated;the contents of ROS,MDA and no in oxidative stress indexes were significantly increased,while the contents of GSH were significantly reduced.Compared with ova,there was no significant change in the measurement indexes of ova + DINP 2 group,while ova + DINP 20 and ova + DINP The levels of Th1 and Th2 cytokines in the 200 groups were significantly decreased,the airway hyperresponsiveness and lung histopathology were significantly increased;the contents of ROS,MDA and NO in oxidative stress indexes were significantly increased,while the contents of GSH were significantly decreased,and the damage to the body was gradually aggravated with the increase of DINP.Conclusion: The above results showed that DINP exposure could aggravate lung tissue injury,airway hyperresponsiveness,promote airway inflammation,and aggravate allergic asthma.The adjuvant effect of DINP may be caused by Th1/Th2 immune system imbalance,and oxidative stress may also participate in it.
