Objective: In this study,SH-SY5 Y cell were used as experimental vectors.Palmitic acid(PA)was used to simulate the high-fat model.SH-SY5 Y cells were pretreated with Rolipram(Rol)to explore the protective effect and mechanism on PA-induced lipotoxicity injury in SH-SY5 Y cells.Research methods: 1.The effect of Rol at different concentrations(0.2,2,20,100,200 ?M)on SH-SY5 Y cells was monitored by MTT,and the optimal Rol concentration was selected.2.MTT was used to monitor the influence of different concentrations of PA(25,50,75,100,150,200,300 ?M)on SH-SY5 Y cell survival rate,and the optimal concentration was selected for modeling.3.According to the experimental results in front of the group are as follows: the Control group,PA(150 ?M),Rol + PA group(Rol10 ?M + PA150 ?M),the Rol group(10 ?M);MTT was used to detect whether Rol had a protective effect on SH-SY5 Y cells under the action of PA.4.DHE(ROS)staining and detection of oxidative stress.5.The expression of apoptotic protein Cleaved caspase-3,Bcl-2 /Bax and signaling pathway related proteins p-MAPK and p-AKT were detected by Western Blot.Results: 1.The survival rate of SH-SY5 Y cells decreased step by step under the action of PA.2.Rol can protect SH-SY5 Y cells by increasing the survival rate of SHSY5 Y cells under the action of PA.3.Rol can inhibit the production of ROS in SHSY5 Y cells under the action of PA,and regulate the apoptotic proteins Cleavedcaspase-3 and Bcl-2 /Bax,suggesting that the protective effect of Rol is related to the inhibition of oxidative stress and apoptosis.4.Rol can increase the level of p-MAPK protein in SH-SY5 Y cells under the action of PA,suggesting that the protective effect of Rol may be related to the MAPK pathway.
