Experimental Study On The Therapeutic Effect Of Yiqi Tongluo Paizhuo Granules On Chronic Prostatitis

Objective: By building model of Chronic prostatitis induced by carrageenan to observe the influence of Yiqi Tongluo Paizhuo Granule on expression of inflammatory cytokines in prostatic tissues,pathological changes in prostatic tissues,and changes in the plasma levels of ET-1,TXB2,6-K-PGF1α and T/K ratio,explore the mechanism of Yiqi Tongluo Paizhuo Granule in the treatment of chronic prostatitis.Methods: 56 healthy male SD rats,SPF grade,4-7 weeks old,were randomly divided into 7 groups,namely blank group,model group,Chinese medicine control group,western medicine control group,high dose group,medium dose group and low dose group.In addition to the blank group,the rest of the groups were intraperitoneally injected with 7% chloral hydrate,after anesthesia to expose the prostate through midline incision,The animal model was established by injecting 0.05 mL of 1% carrageenan saline solution into the dorsal pages of both prostates.After successful modeling,the blank group and model group were gavaged with equal amount of normal saline,the Chinese medicine control group was gavaged with qianqianjiedu capsule solution,the western medicine control group was gavaged with xieerbao capsule solution,and the high,medium and low dose group were gavaged with different concentrations of Yiqi Tongluo Paizhuo Granule solution,for continuous administration for 3 weeks.The rats were anesthetized after administration,blood was collected from the abdominal aorta under aseptic conditions,and the prostate gland was extracted for later use.The plasma levels of ET-1,TXB2,6-K-PGF1α,IL-8,TNF-α were measured by ELISA,and the structures of the prostate tissues stained by HE were observed under microscope.Results:1.Compared with model group,Chinese medicine control group,the western medicine control group,high,medium and low dose group of TNF-α,TXB2 levels drop,T/K ratio decreased;in addition to the western medicine control group,IL-8 levels drop;in addition to the low dose group,ET-1 levels drop;in addition to the traditional Chinese medicine control group and high dose group,6-K-PGF1α levels drop,the difference was statistically significant(P < 0.05).2.Compared with the Chinese medicine control group,the levels of IL-8 and TNF-α in the high,medium and low dose groups were not statistically significant(P > 0.05);Except for medium dose group,ET-1 level increased;TXB2 level decreased in high dose group;6-K-PGF1α level increased in medium dose group and low dose group;and T/K ratio decreased in high dose group and medium dose group,the difference was statistically significant(P < 0.05).3.Compared with the western medicine control group,high,medium and low dose group of IL-8 level difference no statistical significance(P > 0.05);the level of TNF-α in high dose group was obviously lower;high,medium and low dose group of ET-1levels are higher;middle and low dose group of TXB2 levels are higher;high and low dose group of 6-K-PGF1αlevels drop,T/K ratio higher,difference was statistically significant(P < 0.05).Conclusion: Yiqi Tongluo Paizhuo Granule can reduce the expression of inflammatory cytokines in the prostate tissues of rats with chronic prostatitis induced by carrageenan,reduce the plasma hormone ET-1,and restore T/K balance by regulating TXB2 and6-K-PGF1α levels,improve blood circulation,relieve inflammation and local blood stasis symptoms;Medium dose group may be the best dose-effect ratio.
