Purpose:To study the effect of Electroacupuncture on cognitive function and inflammatory cytokines in cerebral cortex of SAMP8 mice,From the perspective of inflammatory response,this paper discusses the effect of Electroacupuncture on the cognitive function of Alzheimer's disease(AD),and discusses its mechanism,so as to provide theoretical guidance for clinical acupuncture treatment of AD.Material and method:SAMP8 of 7-month-old male fast aging model mice and samr1 of anti fast aging model mice were selected,SAMP8 model mice were randomly divided into model group,EA group and western medicine group with 6 mice in each group;SAMR1 model mice were used as the control group,6 mice in each group,all mice are kept in separate cages.In EA group,"Baihui","Shenshu" and "Sanyinjiao" were needled.Continuous wave was selected as electroacupuncture instrument,with frequency of 2 Hz,current of 0.6 m A,lasting for 20 min,and distilled water of the same volume as western medicine was infused into stomach;In the western medicine group,the mice were fixed by intragastric administration of 3 mg / kg / d of doxepin hydrochloride and grabbed once at the same degree;Model group and control group were given the same volume of distilled water by gavage at the same time every day and fixed mice were grabbed once at the same degree.Six days a course of treatment,a total of four courses of treatment.Morris water maze was used to observe the behavior of mice;The distribution and expression of GFAP and Iba1 in astrocytes were detected by immunohistochemistry and Western blot.The distribution and expression of IL-6,IL-1?,TNF-? were detected by immunohistochemistry,Western blot and ELISA.Results:1.The effect of Electroacupuncture on the learning and cognitive ability of SAMP8 mice: In the experiment of positioning navigation,the increase of escape latenct period and swimming distance in the model group was more significant than that in the control group(P < 0.05);the ruduce of escape latent period and swimming distance of EA group and western medicine group was more significant than that in the model group(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).In the space exploration experiment,the reduce of platform crossing number and the target quadrant searching time in the model group was more significant than that in the control group(P <0.05);the increase of platform crossing number and the target quadrant searching time in the EA group and the western medicine group was more significant than that in the model group(P < 0.05);compared with the western medicine group,the number of platform crossing in the EA group was significantly reduced(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).2.The effect of Electroacupuncture on GFAP expression in the cerebral cortex of SAMP8mice: Western blot showed that: the increase of GFAP protein expression in the cortical region in the model group was more significant than that in the control group(P < 0.05);the decrease of GFAP protein expression in the EA group and Western medicine group was more significant than that in the model group(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).3.The effect of Electroacupuncture on the expression of Iba1 in the cerebral cortex of SAMP8mice: ? Immunohistochemical results showed that: the increase of Iba1 positive cells number in the cortical area and the average optical density in the model group was more significant than that in the control group(P < 0.05);the decrease of number and the average optical density in the EA group and Western medicine group was more significant than that in the model group(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).(2)Western blot showed that: the increase of Iba1 protein expression in the cortical region in the model group was more significant than that in the control group(P < 0.05);the decrease of Iba1 protein expression in the EA group and the western medicine group was more significant than that in the model group(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P >0.05).4.The effect of Electroacupuncture on the expression of IL-1 ? in the cerebral cortex of SAMP8 mice: ? Immunohistochemical results showed that: the increase of IL-1? positive cells number in the cortex and the average optical density in the model group was more significant than that in the control group(P < 0.05);the decrease of positive cells number and the average optical density in the EA group and the western medicine group was more significant than that in the model group,(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).(2)Western blot showed that: the increase of IL-1? protein expression in the cortical region in the model group was more significant than that in the control group(P < 0.05);the decrease of IL-1? protein expression in the EA group and the western medicine group was more significant than that in the model group(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).5.The effect of Electroacupuncture on the expression of TNF-? in the cerebral cortex of SAMP8 mice: ?Immunohistochemical results showed that: the increase of TNF-? positive cells number in the cortex and the average optical density in the model group was more significant than that in the control group(P < 0.05);the decrease of positive cells number and the average optical density in the EA group and the western medicine group was more significant than that in the model group,(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).(2)Western blot showed that: the increase of TNF-? protein expression in the cortical region in the model group was more significant than that in the control group(P < 0.05);the decrease of TNF-? protein expression in the EA group and the western medicine group was more significant than that in the model group(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).(3)The results of ELISA showed that: the increase of TNF-? content in the cortical region in the model group was more significant than that in the control group(P < 0.05);the decrease of TNF-? content in the EA group and the western medicine group was more significant than that in the model group(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).6.The effect of Electroacupuncture on the expression of IL-6 in the cerebral cortex of SAMP8mice: in the results of ELISA,the increase of IL-6 content in the cortical region in the model group was more significant than that in the control group(P < 0.05);the decrease of IL-6content in the EA group and the western medicine group was more significant than that in the model group(P < 0.05);there was no significant difference between the EA groups and the western medicine group(P > 0.05).Conclusion:1.The expression of GFAP,Iba1,IL-1 ?,IL-6 and TNF-? in the glial cells of the brain cortex of the aging model mice increased,suggesting that the immune inflammatory response plays an important role in the development of AD;2.Electroacupuncture "Baihui","Shenshu" and "Sanyinjiao" can improve the learning and memory ability of SAMP8 model mice;3.Electroacupuncture "Baihui","Shenshu" and "Sanyinjiao" can improve the learning and memory ability of SAMP8 mice by inhibiting the proliferation and activation of glial cells and the immune inflammatory response.
