Objective: To explore the anticancer effect of Bruceine D on breast cancer,observe its effect on Luminal B human breast cancer cell line MCF-7 cell proliferation,apoptosis and cell cycle,and further explore the mechanism of its apoptosis,so as to provide theoretical basis for clinical medication.Materials : The MCF-7 Cell lines of human breast cancer during logarithmic growth period were used for experiments.The effects of bructin D of different concentrations(0,5,10,20,40,80)on the proliferation of MCF-7 cells were determined by cck-8(Cell Counting kit-8)after 24 h,48h,and 72 h respectively.Flow cytometry was performed to detect the effects of brucocide D on apoptosis and cell cycle of MCF-7 cells for 24 h,48h and 72 h after different concentrations of brucocide(0,10,20,40,80)were used to treat MCF-7 cells for 24 h,48h and 72 h,respectively.Western Blot method was used to detect the expression of JNK and p-JNK proteins in MCF-7 cells treated with bructin D for 24 h.Results: 1.CCk-8 results indicated that the cell survival rate was 98.83%,92.13%,80.38%,68.15%,58.93%,31.25% after 24 h of Bruceine D(0,5,10,20,40,80)?mol/L,respectively.The survival rates of cells at 48 h were 98.12%,85.93%,62.67%,48.75%,30.16% and 23.53%.The cell survival rates of 72 h were 97.73%,71.42%,52.73%,39.96%,26.38%,and 20.13%.2.The results of apoptosis showed that the percentage of apoptotic cells in the total cell population at 24 h was 6.39%,9.18%,14.83%,18.59% and 23.17% after the treatment of(0,10,20,40 and 80)?mol/L of Bruceine D.3.Cell cycle results showed that after 24 h of(0,10,20,40,80)?mol/L of Bruceine D,the proportion of G0/G1 breast cancer cells was 51.45%,58.93%,63.16%,69.38% and 77.38%,respectively.The proportion of s-stage breast cancer cells was35.12%,31.08%,25.32%,22.45% and 18.28%,respectively.The proportion of G2/M stage breast cancer cells was 14.43%,11.99%,9.52%,7.17% and 5.33%,respectively.4.Western Blot results suggested that the expression levels of JNK and p-JNK proteins were up-regulated with the increase of drug concentration after the treatment of(0,10,20,40,80)mol/L bructin D on MCF-7 cells for 24 h.Conclusion:1.The survival rate of MCF-7 cells decreased with the increase of the concentration of Bruceine D,and the longer the duration of the same concentration of the drug,the lower the cell survival rate,showing a significant time-dose dependence(P < 0.05,statistically significant).2.Bruceine D promoted the apoptosis of MCF-7 cells in a time-dose dependent manner,and its proliferation cycle was blocked in the G0/G1 phase.(P < 0.01,statistically significant).3.Bructin D acted on the MCF-7 cell line and up-regulated the expression of JNK and p-JNK proteins,suggesting that bructin D may induce apoptosis of breast cancer MCF-7 cells by activating the JNK signaling pathway and further activating the MAPK signaling pathway.
