Purpose:To investigate the effects of Dioscin from the extract of Dioscorea nipponica on the expression of oxidative stress in the sciatic nerve of mice with painful diabetic peripheral neuropathy and its mechanism of action,to lay a theoretical foundation for the treatment of painful diabetic peripheral neuropathy with a single Chinese medicine,and to find clinical medicine New direction.Material and method:83 male SPF grade C57BL/6 male mice(6-8 weeks old,20-25 g body weight)were selected,and 12 animals were randomly selected as a normal control group after one week of adaptive feeding.The remaining 71 high-fat diets were fed for 8 consecu--tive days after a single intraperitoneal injection of 1% streptozotocin(STZ)70 mg / kg(p H 4.2 in 0.1 mol / L citrate buffer)),and fast for 12 hours before each injection.Blood glucose was measured 72 hours after the second injection,and mice with a blood glucose greater than 16.7 mmol / L proved that the diabetic mouse model was successfully prepared.After 6 weeks,the mechanical pain threshold was measured with Von Frey fiber silk.PDPN mice were modeled when the mechanical pain threshold dropped by more than 50%,and they were randomly divided into a model control group,a low-dose dioscin group,a high-dose dioscin group and ?-lipoic acid group.Each group observed the general condition of mice(including hair color,mental state,food intake,body weight,urine output,daily activities,etc.)and blood glucose at time points of 0 weeks,2 weeks,4 weeks,6 weeks,and 8 weeks.Monitor the pain threshold level of mice at three time points of 0 weeks,4 weeks,and 8 weeks.At 8 weeks,colorimetric method was used to detect the effect of the extract of Dioscorea nipponica on the activity of SOD,GSH-Px and the expression of MDA in sciatic nerve homogenate samples of mice.Results: 1.The mice in the model group had a poor mental state.They suffered from excessive drinking,eating,and polyuria with significant weight loss,bleached and dull coat,and symptoms such as lying on the back and sluggish movements.The mice in the treatment group also had the above symptoms,but Compared with the model group,the degree of improvement in the dioscin high-dose group and ?-lipoic acid group was relatively obvious.2.Compared with the normal group,the blood glucose level of the model group was significantly increased(P<0.01);the blood glucose level of the treatment group was significantly lower than that of the model group at 2 weeks,4 weeks,6 weeks and 8 weeks(P <0.05).3.The body weight level of the model group was significantly lower than that of the normal group(P <0.01);the body weight level of the treatment group was significantly higher than that of the model group at 4 weeks,6 weeks,and 8 weeks(P<0.01).4.The levels of thermal pain threshold and mechanical pain threshold of the model group were significantly lower than those of the normal group(P<0.01);compared with the model group,the heat pain threshold and mechanical pain threshold level in the treatment group at 4 and 8 weeks were significantly higher(P<0.01).5.At 8 weeks,the SOD,GSH-Px enzyme activity and MDA levels in the sciatic nerve tissue homogenate of mice in each group were compared: The level of SOD in the sciatic nerve tissue homogenate of the model group and the low-dose dioscin group was significantly lower than that of the normal group(P<0.01),and the level of SOD in the sciatic nerve tissue homogenate of the high-dose dioscin group and alpha-lipoic acid group Lower than normal group but the difference is not statistically significant(P>0.05).The level of SOD in the sciatic nerve homogenate of mice in the low-dose dioscin group,high-dose dioscin group,and alpha-lipoic acid group was significantly higher than that in the model group(P<0.05).The level of GSH-Px enzyme activity in the sciatic nerve tissue homogenate of the model group,the low-dose dioscin group,the high-dose dioscin group and the alpha-lipoic acid group was significantly lower than that of the normal group(P<0.05).The level of GSH-Px enzyme activity in the sciatic nerve tissue homogenate of mice in the low-dose dioscin group,high-dose dioscin group,and alpha-lipoic acid group was significantly higher than that in the model group(P<0.01).MDA levels in sciatic nerve tissue homogenate of model group and low-dose dioscin mice were significantly higher than normal group(P<0.05),MDA levels in sciatic nerve tissue homogenate of mice in high-dose dioscin group and alpha-lipoic acid group higher than the normal group but the difference was not statistically significant(P>0.05).MDA levels in the sciatic nerve tissue homogenate of mice in the low-dose dioscin group,high-dose dioscin group,and alpha-lipoic acid group were significantly lower than that in the model group(P<0.01).Conclusion: Dioscin from the extract of Dioscorea nipponica can inhibit the occurrence and development of oxidative stress,reduce the pain symptoms of mice with painful diabetic peripheral neuropathy,and improve the nerve damage of mice with painful diabetic peripheral neuropathy.
