The interference of potassium ion homeostasis is one of the ways to cause cell dysfunction.Crown ether is an excellent ion carrier that can transport potassium ions and has great potential for biomedical applications.In this paper,three amphiphilic crown ether micelles were prepared and their inhibitory effects on cancer cell activity were investigated.The main research work and results of this paper are as follows:Crown ether-PEG?polyethylene glycol?-PCL?poly caprolactone?micelles?CPMs?were prepared by esterification and ring-opening reaction with hydroxyl-PEG-carboxyl(HO-PEG5000-COOH),crown ether and caprolactone as raw materials.The experimental test showed that the hydration particle size of the micelles was about120nm,and the particle size was about 50nm under TEM observed.The particle size was stable within 7 days,no significant changes were observed after incubation with BSA solution,and the micelle concentration?CMC?was low.It has a certain complexation for potassium ion,and with the increase of concentration,the complexation of CPMs becomes more obvious.Subsequent anti-tumor experiments:MTT results showed that CPMs showed no obvious toxicity when the concentration increased from 31.25?g/mL to 125?g/m L With the continuous increase of the concentration,the cytotoxicity increased at 250?g/mL,and CPMs significantly inhibited the growth of HeLa cells when the concentration reached 500?g/mL.At the same time,CPMs could be absorbed by HeLa cells at 2 h by laser confocal observation.Next,in vivo anti-tumor experiments of CPMs also produced certain effects,and no obvious adverse reactions were observed.The final histological analysis also demonstrated a therapeutic effect on the tumor without significant side effects.In summary,a kind of micelle with crown ether was prepared in this study.The micelle had good stability,certain complexation effect on potassium ion,certain inhibition effect on HeLa cell and tumor growth,and no obvious side effects.Different from traditional drug chemotherapy,crown ether has shown the potential of biomedicine,and the interference of ion homeostasis has the potential to become a new way of tumor treatment. |
