| Objective:To test the effects of Lipi compound preparation on LP,NPY and CCK-8,the brain-gut peptide,in normal gastric antrum smooth muscle cells and gastric antrum tissue of anorexia rat,exploring the mechanism and specific targets of Lipi compound preparation in treating anorexia,and to further reveal the theoretical basis and scientific basis of Lipi compound preparation in preventing and treating infantile anorexia.Methods:(1).60 rats were selected,sex in half,were randomly divided into six groups,10 rats in each group,named:cell culture group,saline group,LiPi compound preparation of low,middle and high dose group and zinc sulphate group.Cell culture group was used for isolation and culture of primary rat gastric antral smooth muscle cells.The other groups were given normal saline,low,medium and high doses of Lipi compound preparation and zinc sulfate solution,once a day,for 7 consecutive days,after the last gastric perfusion for 1 hour,to prepare drug-containing serum.10%serum was used to intervene rat gastric antrum smooth muscle cells to find the optimal time for cell proliferation.The expression of LP,NPY and CCK-8 in rat gastric antrum smooth muscle cells was detected by RT-PCR and Western blot at the optimal time point for cell proliferation.(2).Thereafter,50 rats in the model group,sex in half,with,10 rats in each group,were randomly divided into model control group,low,medium and high dose of Lipi compound preparation group and zinc sulfate group.Normal group and model control group were given normal saline,low,medium and high dose groups were given low,medium and high dose Lipi compound preparation.Zinc sulfate solution was given to zinc sulfate group once a day for 7 consecutive days.Antrum tissues were taken after the last gastric perfusion for 1 hour.The expression of LP,NPY and CCK-8 in rat gastric antrum tissue was detected by Western blot and Elisa.Result:1.The serum containing drugs in each group could promote the proliferation of rat gastric antrum smooth muscle cells.The optimum time for cell proliferation was 12 hours.The serum containing drugs in high dose Lipi compound preparation was the best.2.Compared with saline group,different concentrations of Lipi compound preparation and zinc sulfate containing serum could up-regulate the expression of LP and NPY in rat gastric antrum smooth muscle cells,and down-regulate the expression of CCK-8.The serum containing zinc sulfate had the best effect,and the high dose group had the best effect in different concentration of Lipi Compound Preparations.3.After the successful establishment of anorexia rat model,the intake of anorexia rats was 52.2%lower than that of normal rats,and the weight of anorexia rats was 26.1%lower than that of normal rats.After drug intervention,the food intake and body weight of each group were restored to varying degrees.Zinc sulfate had the best restoring effect,and the high dose group had the best effect in different concentration of Lipi compound preparation.4.Compared with the normal group,the expression of LP,NPY and CCK-8 protein in gastric antrum tissue of anorexic rats decreased,and the expression of LP,NPY and CCK-8 protein was up-regulated by drug intervention.Zinc sulfate had the best effect in each treatment group,and the medium dose group had the best effect in different concentration of Lipi compound preparation.Conclusion:1.In physiological state,LP,NPY and CCK-8 were expressed in rat antral smooth muscle cells.Lipi Compound and Zinc Sulfate Serum have regulatory effects on the expression of three brain-gut peptides,including up-regulation of LP,NPY and down-regulation of CCK-8,in order to optimize the regulation of Lipi high-dose group.This may be one of the mechanisms of Lipi Compound in the treatment of anorexia by regulating the changes of brain intestinal peptide in gastric antrum smooth muscle cells,promoting the feeding behavior of appetite center and exerting its "invigorating spleen and promoting transportation" effect.2.The expression of LP,NPY and CCK-8 in gastric antrum tissue of anorexia rats was decreased under pathological condition.The disorder of the expression of three brain-gut peptides may be related to the occurrence of anorexia.The expression of LP,NPY and CCK-8 in gastric antrum tissue was up-regulated after the intervention of Manager Spleen Compound Preparations and Zinc Sulfate,and the middle dose group was the best.This may be one of the mechanisms of Lipi Compound in treating anorexia. |
