EXPRESSION OF HOTAIR,LC3 AND P62 IN HUMAN NUCLEUS PULPOSUSObjective To analyze the expression of HOTAIR and autophagy markers LC3 and p62 in the nucleus pulposus of intervertebral disc and their relationship with the degree of intervertebral disc degeneration.MethodsThe expression levels of HOTAIR and autophagy markers(LC3-I,LC3-II and p62)in the nucleus pulposus tissues of 30 patients(6 Cases of Pfirrmann ?,6 Cases of Pfirrmann III,10 Cases of Pfirrmann IV and 8 Cases of Pfirrmann V)were detected by Quantitative real time polymerase chain reaction(qRT-PCR).Analyze the correlation between the expressions of HOTAIR,p62 and LC3-II/I ratio and the degree of disc degeneration.ResultsThe qRT-PCR test showed that the expression of HOTAIR gradually decreased with the increase of disc degeneration grade,and was negatively correlated with the degree of disc degeneration(r=-0.67,P<0.05).With the increase of intervertebral disc degeneration grade,LC3-II/I ratio also gradually decreased,and was negatively correlated with intervertebral disc degeneration grade(r=-0.79,P<0.05),and the expression of p62 gradually increased,and was positively correlated with intervertebral disc degeneration grade(r=0.46,P<0.05).ConclusionThe degree of degeneration of human intervertebral disc was negatively correlated with the expression of HOTAIR and LC3-?/I ratio,and positively correlated with the expression of p62.EXPLORES THE EFFECTS OF HOTAIR ON THE EXPRESSION OF LC3 AND P62 IN HUMAN NORMAL NUCLEUS PULPOSUS CELLSObjective To investigate whether HOTAIR is correlated with LC3-II/I ratio and p62 expression in human normal nucleus pulposus cells.Methods Human normal nucleus pulposus cells were treated with liposomes transient transfection method to construct si-NC group,si-HOTAIR group,Vector-NC group,and Vector-HOTAIR group.The expression of LC3-I,LC3-II,and p62 in nucleus pulposus cells were detected by q RT-PCR and WB.The changes of the expression of p62 and LC3-II/I ratio after the inhibition and overexpression of HOTAIR were observed.Results qRT-PCR and WB tests showed that the ratio of LC3-II/I was significantly lower than that of the control group(P<0.01)and the expression of p62 was significantly higher than that of the control group after the HOTAIR gene was repressed(P<0.01).The LC3-II/I ratio was significantly higher after overexpression of HOTAIR gene than that of the control group(P<0.01),while the expression of p62 was significantly lower than that of the control group(P<0.01).Conclusion HOTAIR can affect autophagy of human nucleus pulposus cells by up regulating the expression of LC3-II / I ratio and down regulating p62 expression.
