Objective:To explore the possible mechanism of Zhuangdu Quhan Mixture(ZQM)in treating ankylosing spondylitis by investigating the effects of ZQM on arthritis symptoms,miR-29 a and Wnt signaling pathway related factors in PGIA mice.Methods:Fifty female BALB/c mice were randomly divided into normal group,model group,western medicine group,low-dose ZQM group and high-dose ZQM group(n=10 per group).Except for the normal group,the others were injected of complete Freund's adjuvant plus proteoglycan intraperitoneally to replicate the PGIA mice.Two weeks after the last immunization,the corresponding drug treatment began.The AI scores were measured every week.The expressions of miR-29 a,DKK1 mRNA and?-catenin mRNA were determined by quantitative Real-time PCR,and the protein contents of DKK1 and ?-catenin were detected by Western blotting.Results:1.The effect of ZQM on PGIA mice:ZQM can effectively reduce the arthritis symptoms of PGIA mice.Compared with the model group,the AI score of mice in each treatment group decreased(P<0.05),There was no significant difference between high-dose ZQM group ? low-dose ZQM group and western medicine group(P>0.05).2.The effect of ZQM on miR-29a: Compared with the model group,the expression of miR-29 a in each treatment group decreased significantly(P<0.01).Compared with low-dose ZQM group and western medicine group,the expression of miR-29 a in high-dose ZQM group decreased significantly(P<0.01).3.The effect of ZQM on factors related to Wnt pathway: Compared with the model group,the expression of DKK1 mRNA and protein in each treatment groups increased(P<0.05 or p<0.01),and the expression of?-catenin mRNA and protein decreased(P<0.05 or p<0.01).Compared with the low-dose ZQM group and the western medicine group,the expression of DKK1 mRNA in the high-dose ZQM group increased(P<0.01),the expression of DKK1 protein was not statistically significant(P>0.05),and the expression of ?-catenin DKK1 mRNA and protein decreased(P<0.05 or p<0.01).Conclusion:1.ZQM can alleviate the arthritis symptoms of PGIA mice and reduce the AI score.2.ZQM can inhibit the expression of miR-29 a and ?-catenin,and activation the expression of DKK1,suggesting that ZQM may delay the formation of new bone in AS by regulating Wnt/?-catenin signaling pathway through miR-29 a.
