Objective: To observe and discuss the relationship between "drinking blood stasis and treating the same method" on pulmonary vascular remodeling in rats with hypoxic pulmonary hypertension,aiming at the pathogenesis of HPH,exploring effective prevention and treatment methods,and providing a certain experimental basis for clinical application.Methods: Under normal pressure and hypoxia conditions,the rat model of hypoxic pulmonary hypertension was reproduced according to Peng Gongyong and other methods.Fifty healthy male SD rats were divided into normal group,HPH model group,prednisone group,traditional Chinese medicine and high-dose group,10 rats in each group were placed in hypoxic chamber for 8 hours after daily administration.After 21 days of continuous intervention,m PAP was measured and recorded according to Sun Bo et al.'s modified right heart catheterization.The lung tissues of the same part were taken and stained with HE andelastic fibers to observe the changes of pulmonary vascular pathology.CTGF and TGF-?1 m RNA expression and IOD values of rat lung tissue were detected. result:1.After 1 week of hypoxia,the model-building rats showed poor mental state,messy hair,slight breathing,increased secretions from the nose and mouth,less movement and less food,indicating that the general conditions and food intake of rats under hypoxic conditions affected.2.Compared with the normal group,the m PAP of the HPH model group was significantly increased,and the difference was statistically significant(P<0.05),indicating that the hypoxic pulmonary hypertension rat model was successfully replicated.3.HE staining and elastic fiber staining: The pulmonary vascular wallof the HPH model group was significantly thickened,the medial smooth muscle cells proliferated,the lumen stenosis was obvious,the endothelial cells were continuously destroyed,and the elastic fiber hyperplasia and damagewere seen in the lumen,The characteristics of pulmonary vascular remodeling appear.4.CTGF and TGF-?1 m RNA expression in rat lung tissue: Compared with the HPH model group,the normal group,prednisone group,Chinese medicine medium and high dose groups all decreased,the difference was statistically significant(P<0.05).5.IOD values of CTGF and TGF-?1 expression in rat lung tissue: Compared with the HPH model group,the normal group,Chinese medicine,high-dose group were reduced,the difference was statistically significant(P<0.05).conclusion:1.Hypoxic modeling caused an increase in the average pulmonary artery pressure in rats."Tongyin Tongzi" decoction can reduce the average pulmonary artery pressure in rats with hypoxic pulmonary hypertension.2.Hypoxia can cause thickening of the pulmonary vascular wall,narrowing of the lumen,and the appearance of pulmonary vascular remodeling;Compound Tinglizi Decoction of "Drinking Blood Stasis and Tongzhi" can down-regulate the expression of CTGF and TGF-?1 m RNA in rat lung tissue.It can inhibit or even partially reverse pulmonary vascular remodeling.
