| Objective:To observe the effect of electroacupuncture on BWT score of rats in focal cerebral ischemia model,and to clarify the regulatory effect of electroacupuncture on NR1 and Ca2+.Methods:90 SPF grade SD rats were randomly divided into normal control group(18),sham operation control group(18)and model group(54)according to the random number table method.The MCAO rat model on the left side was prepared by improved thread bolting method reported by Zea Longa and Knaga.After successful modeling,the modeling grou P was randomly divided into model control group,electroacupuncture pericardial acupoint group and electroacupuncture pulmonary acupoint group.The pericardial acupoint group and the pulmonary acupoint group received electroacupuncture treatment once a day for 3 consecutive days respectively on the second day after successful modeling.After binding,the rats were injected with no.15mm and no.36 filiform needles into the paralyzed side acupuncture point of the rats,with a depth of 3-4mm.The pericardium was divided into one group by tianquan and quze acupoints,and the other group by neiguan and daling.Lung by tianfu,chi ze a group,lieque,taiyuan a group.The stimulation parameters of electroacupuncture are continuous wave(sparse wave),the output current is 0.5-1m A,the frequency is2-15Hz,the intensity is light tremor of local tissue,and the continuous stimulation time is 30min.Other groups were fixed with the same method for 3 days.The changes of BWT score before and after treatment were observed in each group.TTC staining,immunohistochemistry and flow method were used to detect the volume of infarcted brain tissue and the concentration of NR1 and Ca2+in brain tissue.Results:1.Comparison of general conditions of rats:there was no significant difference in general conditions between the normal group and the sham operation group,all of which showed good mental state,shiny fur and normal diet.The rats in the model group were in poor mental state,with hair loss,sombre,no desire to eat,irritability,and significantly reduced limb strength on the affected side.Compared with the model group,the lung meridian group and pericardial meridian group had slightly shiny hair,and the feeding condition,mental state and limb strength of the affected side were all stronger than those of the model group.2.Comparison of BWT scores in rats:(1)comparison between groups:before treatment:compared with the normal group and the sham operation group,the scores of model group,pulmonary meridian group and pericardial meridian group were all decreased,the differences were statistically significant(P<0.05).There was no significant difference in the ratio between the three groups(P>0.05).After treatment:compared with the normal group,the sham operation group and the model group,the pericardial meridian group showed an increase in scores,the difference was statistically significant(P<0.05),and there was no statistical difference with the lung meridian group(P>0.05).(2)intra-group comparison:compared with that before treatment,the scores of pulmonary meridian group and pericardial meridian group were all increased,the differences were statistically significant(P<0.05).3.Comparison of cerebral infarction volume:compared with the normal group and the sham operation group,the volume of the model group,the pulmonary meridian group and the pericardial meridian group all increased,showing statistical significance(P>0.05).MCAO modeling success;There was no statistically significant difference between the model group,the pulmonary meridian group and the pericardial meridian group(P>0.05).4.NR1 expression:compared with the normal group,NR1 positive expression was increased in the model group,the lung meridian group and the pericardial meridian group,showing statistical significance(P<0.05).Compared with the sham operation group,the positive expressions in the model group and the lung group were all increased,with statistical significance(P<0.05),and the difference in the pericardium group was not statistically significant(P>0.05).Compared with the model group,the expression of NR1 in the pericardium group was significantly decreased,which was statistically significant(P<0.05).5.Ca2+content:Compared with the normal group and the sham operation group,the contents of the model group,the pulmonary meridian group and the pericardial meridian group were all increased,and the differences were statistically significant(P<0.05).(2)Compared with the model group,the contents of both pulmonary and pericardial meridional groups decreased,with statistically significant difference(P<0.05).Conclusion:1.Electroacupuncture treatment of pericardial meridian can improve the symptoms of nerve functional defects in MCAO rats,but there is no significant change in the volume of cerebral infarction.2.Cerebral ischemia can lead to enhanced NR1 expression and significantly increased Ca2+concentration in MCAO rats.Electroacupuncture at pericardial acupoint can down-regulate NR1 expression and reduce Ca2+concentration. |
