Purpose:To explore the change of medicine property before and after the compatibility of Paeoniae Alba Radix and Glycyrrhizae Radix based on the theory of traditional Chinese medicine property.Research the secondary metabolite content change and the primary material content change in the compatibility of Paeoniae Alba Radix and Glycyrrhizae Radix before and after mixing to argue the rationality,then to research the effects on body energy metabolism.Material and method:1.The content of protein was determined by Coomassie Brilliant Blue method;Polysaccarides was determined by Phenol sulfate acid method;Using Soxhlet extraction and oven drying method to determine the content of lipids in samples and moisture content.Use SPSS 17.0 and analyzed by Kruskal-Wallis test and t test.2.Use central composite design,Independent variables were solvent concentration,the time of extraction.ratio of solvent,The proper extracting condition was optimized with the amount of albiflorin,paeoniflorin,liquiritin apioside,liquritin as index.Use HPLC to optimize extraction process.Hopping to offer the reference to explain the theory of traditional Chinese medicine property.3.Adopting the diet restriction method and high-protein diet to establish the weak model and the strong model;divided the mice into blank group,weak model group,weak model group given Paeoniae Alba Radix,weak model group given Glycyrrhizae Radix,weak model group given drug pair,strong model group,strong model group given Paeoniae Alba Radix,strong model group given Glycyrrhizae Radix,strong model group given drug pair recording the ratio of the time that mice stay in the cold/hot plate after given Glycyrrhizae Radix,Paeoniae Alba Radix and the drug-pair extracts according to the tendency behavior of temperature using the cold/hot plate differentiating technology to exploring the transform of the medicinal properties of Glycyrrhizae Radix,Paeoniae Alba Radix and the drug-pair being from mice metabolism combined with some energy indexes(the oxygen consumption,Na+K+-ATPase,Ca2+Mg2+-ATPase),antioxidation indexes(T-AOC,SOD),blood indexes(NE,DA)and the body temperature,the biochemical index measured by kit method.Results:1.The statistical results represent that there are significant differences among Glycyrrhizae Radix,Paeoniae Alba Radix before and after they paired(P<0.05);There are significant differences between Glycyrrhizae Radix and the drug-pair(P<0.05);There are significant differences between Paeoniae Alba Radix and the drug-pair(P<0.05).2.The proper extracting condition was methanol concentration was 80 per-cent,and extracted with 300 fold methanol ultrasound extraction for 30min.3.aiming at the analyzation of NE、DA、Na+K+-ATPase、Ca2+Mg2+-ATPase、T-AOC、SOD and the body temperature、the oxygen consumption and the time of staying in the hot pad,it shows that Paeoniae Alba Radix could decrease the oxygen consumption,Na+K+-ATPase,Ca2+Mg2+-ATPase,T-AOC,SOD,NE,DA and the body temperature,increase the ratio of staying in the hot pad.Nevertheless,Glycyrrhizae Radix gives the opposite effect.The effect of the drug-pair could decrease the ratio of staying in the hot pad,increase the oxygen consumption,temperature,and the Na+K+-ATPase,Ca2+Mg2+-ATPase,T-AOC,SOD,NE and DA,but no obvious influence on the Glycyrrhizae Radix.Conclusion:The degree of medicinal property tended to ease and finally it manifested the‘heat nature’.
